Anxiety disorders are a leading source of human misery, morbidity, and premature mortality. Existing treatments far from curative for many, underscoring the need to clarify underlying neural mechanisms. Although many brain regions contribute, amygdala has received most intense scientific attention. Over past several decades, this scrutiny yielded detailed understanding function, but it failed produce new clinical assays, biomarkers, or cures. Rising urgent public health challenge demands an honest reckoning with functional-neuroanatomical complexity shift theories anchored on "the amygdala" models centered specific nuclei cell types. This review begins by examining evidence studies rodents, monkeys, humans "canonical model," idea that plays central role in fear- anxiety-related states, traits, disorders. Next, authors selectively highlight work indicating canonical model, while true, is overly simplistic fails adequately capture actual state evidentiary record, breadth amygdala-associated functions illnesses, amygdala's functional architecture. The describe implications these facts basic neuroimaging research. concludes some general recommendations grappling accelerating efforts understand more effectively treat amygdala-related psychopathology.

Abstract Exploring the neurobiological effects of various treatments for depression is pivotal in elucidating shared and unique mechanisms action that may explain commonalities differences treatment outcomes. The amygdala, consisting subdivisions with distinct roles emotional processing, thought to be implicated pathophysiology depression. However, how alter nuclei-specific resting-state functional connectivity (rs-FC) amygdala remains unclear. Here, we hypothesized restore rs-FC abnormalities changes. To test this hypothesis, multicenter observational study analyzed MRI data patients (N = 187) healthy controls 113). Patients were scheduled undergo either cognitive behavioral therapy (CBT; N 63), medication 43), electroconvulsive (ECT; 32), or transcranial magnetic stimulation (TMS; 26). Nine subregions delineated using FreeSurfer segmentation module. enhance interpretability reliability, these further clustered by applying k-means + clustering time courses, resulting three clusters referred as basolateral (BL), basomedial (BM), centromedial (CM) based on their anatomical locations. At baseline, BL BM exhibited hyperconnectivity mediodorsal thalamus (MDTH), periaqueductal gray, perigenual anterior cingulate cortex, dorsal raphe nucleus, dorsomedial prefrontal cortex. Following treatment, there no changes across types; however, BL-MDTH decreased level following ECT. These results demonstrate ECT-specific restoration exists while multiple do not share common amygdala.

Abstract Neuroimaging research has yet to elucidate whether reported gray matter volume (GMV) alterations in major depressive disorder (MDD) exist already before the onset of first episode. Recruitment presently healthy individuals with a subsequent transition MDD (converters) is extremely challenging but crucial gain insights into neurobiological vulnerability. Hence, we compared converters patients and sustained controls (HC) distinguish pre-existing markers from those emerging later course depression. Combining two clinical cohorts ( n = 1709), voxel-based morphometry was utilized analyze GMV 45 converters, 748 MDD, 916 HC region-of-interest approach exploratory whole-brain. By contrasting subgroups considering both remission state recurrence at 2-year follow-up, stepwise disentangled effects (1) vulnerability, (2) acute state, (3) an initial vs. recurrent Analyses revealed higher amygdala relative p tfce-FWE 0.037, d 0.447) 0.005, 0.508), remaining significant when remitted imminent recurrence. Lower dorsolateral prefrontal cortex < 0.001, 0.188) insula 0.010, 0.186) emerged not driven by MDD. examining one largest available converter samples psychiatric neuroimaging, this study allowed determination neural for impending Our findings suggest temporary which combination other common risk factors might facilitate prediction turn improve prevention

The amygdala plays a role in behavior and emotional response is vulnerable to Alzheimer's disease (AD) pathology, yet little known about tau accumulation before clinical symptom onset. To investigate whether certain nuclei are particularly degeneration might underlie early neuropsychiatric symptoms AD, we aimed characterize subregional pathology its correlates associations with established biomarkers of AD cognitive-behavioral measures Presenilin-1 E280A mutation carriers autosomal dominant AD. Participants included 25 cognitively unimpaired 37 non-carrier family members from the Colombia-Boston (COLBOS) Biomarker Study. Measures 18F-flortaucipir, 11C-Pittsburgh compound B, Consortium Establish Registry for Disease Word List Learning, Trail Making Test, Geriatric Depression Scale, Anxiety Inventory. We examined group differences levels (whole amygdala, lateral nucleus basal nucleus) analyzed markers measures. Amygdala were higher compared non-carriers. Among carriers, showed greater burden than nucleus, correlated closer estimated age onset increased cortical amyloid. Additionally, both was associated poorer working memory, lower executive function depressive symptoms. However, did not correlate anxiety. Notably, differentiated non-carriers, predictive accuracy when included. These findings suggest that begins while cognitive deficits nuclei's differential vulnerability underscores importance investigating spread within amygdala-associated networks, relative manifestations This study reinforces potential as valuable biomarker preclinical
