Insecticide resistance has recently become a serious problem in the agricultural field. Development of insecticides with new mechanisms action is essential to overcome this limitation. Juvenile hormone (JH) an insect-specific that plays key roles maintaining larval stage insects. Hence, JH signaling pathway considered suitable target development novel insecticides; however, only few inhibitors (JHSIs) have been reported, and no practical JHSIs developed. Here, we established high-throughput screening (HTS) system for exploration using Bombyx mori cell line (BmN_JF&AR cells) carried out large-scale chemical library. The four-step HTS yielded 69 compounds as candidate JHSIs. Topical application JHSI48 B. larvae caused precocious metamorphosis. In ex vivo culture epidermis, suppressed expression Krüppel homolog 1 gene, which directly activated by JH-liganded receptor. Moreover, parallel rightward shift response curve, suggesting possesses competitive antagonist-like activity. Thus, libraries may applications future targeting pathway.

The yellow fever mosquito, Aedes aegypti, vectors human pathogens. Juvenile hormones (JH) control almost every aspect of an insect's life, and JH analogs are currently used to mosquito larvae. Since RNA interference does not work efficiently during the larval stages this insect, regulation development mode action well studied. Abstract BACKGROUND Intensive application of acetolactate synthase (ALS)‐inhibiting herbicides has resulted in herbicide‐resistance many weeds, including Bromus sterilis . The present study was conducted to identify the mechanisms conferring resistance ALS‐inhibiting a biotype. RESULTS Dose–response studies revealed resistant biotype be 288 times less sensitive pyroxsulam than susceptible Furthermore, experiment with single‐dose, proved this also cross‐resistant propoxycarbazone, iodosulfuron plus mesosulfuron and sulfosulfuron. Prior treatment malathion, known inhibitor cytochrome P450s, reduced level pyroxsulam. No mutations were detected from partial ALS gene sequencing. Flow cytometry chromosome counting rejected ploidy variation between biotypes. Relative copy number ruled out amplification. Abstract The monarch butterfly ( Danaus plexippus ) complements its iconic migration with diapause, a hormonally controlled developmental programme that contributes to winter survival at overwintering sites.
