Hu sheep, an indigenous breed in China known for its high fecundity, are being studied to improve their growth and carcass traits. MSTN is a negative regulator of muscle development, inactivation results muscularity. The C-CRISPR system, utilizing multiple neighboring sgRNAs targeting key exon, has been successfully used generate genes complete knockout (KO) monkeys mice one step. In this study, the system was MSTN-edited sheep; 70 embryos injected with Cas9 mRNA four exon 3 sheep were transferred 13 recipients. Out 10 lambs born from five recipients after full-term pregnancies, nine had KO various mutations. No off-target effects found. These MSTN-KO showed double-muscled (DM) phenotype, characterized by higher body weight at 4 months old, prominent muscular protrusion, clearly visible intermuscular groves, hypertrophy. molecular analysis indicated enhanced AKT suppressed ERK1/2 signaling gluteus edited sheep. conclusion, DM phenotype efficiently specifically generated using C-CRISPR, method promising tool farm animal breeding.

How to increase meat production is one of the main questions in animal breeding. Selection for improved body weight has been made and, due recent genomic advances, naturally occurring variants that are responsible controlling economically relevant phenotypes have revealed. The myostatin (MSTN) gene, a superstar gene breeding, was discovered as negative controller muscle mass. In some livestock species, natural mutations MSTN could generate agriculturally desirable double-muscling phenotype. However, other species or breeds lack these variants. 绵羊的体重大小能直接反映羊只的生长速度、肥育速度, 也是衡量肉羊生长性能的重要指标. 本研究以乌珠穆沁羊为研究对象. 挑选出100个与绵羊体型相关的SNP位点, 同时采集252只乌珠穆沁羊的血液和不同时期体重数据, 使用MassARRAY基因分型技术对这些绵羊在选定的SNP位点进行基因分型, 并将体重数据与基因分型结果进行关联分析. 结果表明, 本研究共验证出5个突变位点与乌珠穆沁羊不同时期体重显著相关. 进一步对3个错义突变位点进行蛋白结构预测分析发现, 3个突变位点CDS序列改变导致其蛋白质三级结构发生不同程度改变, 进而导致个体体重差异. 本研究旨在挖掘与乌珠穆沁羊生长性状相关的特征位点, 为乌珠穆沁羊今后的精准选育提供参考, 以期推动乌珠穆沁羊产业的发展.
