Fatty acid metabolism disruptions affect low-grade gliomas (LGGs), with glioma cells depending on fatty acids for survival. Targeting oxidation through the acyl-coenzyme A synthetase long-chain (ACSL) family could alleviate growth and improve prognosis management. However, integration of ACSLs analyzing their relationship LGGs remains unexplored. We collected RNA expression data from TCGA, GTEx, CGGA, GEO datasets validated prognostic significance gene in 37 samples. DNA methylation UCSC Xena promoter levels via MEXPRESS were analyzed. Functional enrichments co-expressed genes conducted using Gene Ontology, Kyoto Encyclopedia Genes Genomes, Set Enrichment Analysis. Protein–protein interaction networks established GeneMANIA, cBioPortal assessed somatic mutations copy number variations LGGs. TIMER TISIDB databases investigated correlation between immune infiltration checkpoint genes. Hazard ratios (HR) 95% confidence intervals (95% CI) computed, net reclassification index integrated discrimination improvement estimated to evaluate predictive capability model. Independent factors overall survival included age, gender, tumor grade, MGMT status, ACSL1, ACSL3, ACSL5, ACSL6 levels. High ACSL1 (HR = 2.352, 95%CI: 1.647–3.359, P 9.00E-06), ACSL3 2.367, 1.547–3.624, 2.92E-04) ACSL5 2.329, 1.611–3.367, 2.80E-05) correlated poor prognosis, while increased 0.449, 0.290–0.696, 1.02E-03) related better rates. Furthermore, these associations also confirmed validation our external cohort. Negative was found. enrichment analyses highlighted roles mechanisms function, significant CNVs cell infiltration. exhibit influence immunity migration, providing valuable insights into LGG therapeutic targets.
