Antarctic krill (Euphausia superba), which is rich in astaxanthin, has been widely utilized as a dietary supplement fish aquaculture. Our study was to feed juvenile leopard coral grouper (Plectropomus leopardus) diet containing 50 % krill, revealing significant body color differentiation between reddened group (BKR) and non-reddened (BKB), followed by comparative analysis with the control (BCon) without supplementation. Histological carotenoid content liver intestine were differentially regulated color-differentiated individuals. Transcriptomic profiling revealed expressed genes (DEGs) among individuals, up-regulated DEGs BKR being linked uptake, metabolism, transport. Key (acss2l, insig1, fabp7, bco1) validated through qRT-PCR FISH. Additionally, WGCNA identified potential gene regulatory networks that responsive coloration. The leopard coral grouper (Plectropomus leopardus) is a commercially significant tropical marine species. With the ongoing effects of global climate change, increasing attention has been focused on grouper's susceptibility to extreme cold weather. This study investigates acute exposure and temperature recovery liver P. leopardus. Histological observations enzyme activity assays revealed that fluctuations caused disruptions normal physiology, including lipid accumulation alterations in antioxidant levels. Transcriptomic analysis tissue identified 2744 differentially expressed genes (DEGs) across three experimental groups: 25 °C (control), 13 (cold exposure), rewarming at (R-25 °C). Functional enrichment these DEGs were significantly associated with biological processes such as metabolism defense, well pathways related metabolism, fatty acid biosynthesis, ferroptosis. Furthermore, dynamic regulation immune responses, oxidative stress was observed response both rewarming. Notably, several redox-related identified, their interactions further explored.
