Causal effects of nonalcoholic fatty liver disease on cerebral cortical structure: a Mendelian randomization analysis DOI Creative Commons

Background Previous studies have highlighted changes in the cerebral cortical structure and cognitive function among nonalcoholic fatty liver disease (NAFLD) patients. However, impact of NAFLD on specific affected brain regions remains unclear. Therefore, we aimed to explore potential causal relationship between structure. Methods We conducted a Mendelian randomization (MR) study using genetic predictors alanine aminotransferase (ALT), NAFLD, percent fat (PLF) combined them with genome-wide association (GWAS) summary statistics from ENIGMA Consortium. Several methods were used assess effect full cortex regions, along sensitivity analyses. Results At global level, PLF nominally decreased SA cortex; at functional ALT presented nominal reduced parahippocampal gyrus, TH pars opercularis, orbitalis, pericalcarine cortex. Besides, triangularis cortex, but increased entorhinal lateral orbitofrontal temporal pole. Furthermore, cuneus lingual Conclusion is suggestively associated atrophy human brain.
