Tumor immune microenvironment is crucial for diffuse large B-cell lymphoma (DLBCL) development. However, the mechanisms by which super-enhancers (SEs) regulate interactions between DLBCL cells and tumor-infiltrating remains largely unknown. This study aimed to investigate role of SE-controlled genes in regulating cells. Single-cell RNA-seq, bulk RNA-seq H3K27ac ChIP-seq data were downloaded from Heidelberg Open Research Data database Gene Expression Omnibus database. HOMER algorithm Seurat package R used bioinformatics analysis. Cell proliferation lactate dehydrogenase (LDH) release was detected MTS LDH assays, respectively. Interaction B cell cluster CD8 + T NK most obviously enhanced DLBCL, with CD70-CD27, MIF-CD74/CXCR2 complex, MIF-CD74/CD44 complex CCL3-CCR5 significantly increased. sub-cluster showed strongest interaction cluster. ZZZ3 upregulated transcription CD70 binding its SE. Silencing DOHH2 promoted co-cultured NK92 cells, counteracted overexpression silencing combined PD-L1 blockade In conclusion, inhibited killing infiltrating ZZZ3/CD70 axis. Targeting axis expected be a promising strategy treatment.

Negative factors present in TME, such as IL-10, may limit effectiveness cellular vaccines based on dendritic cells, therefore, it is important to control its effect. influence IL-10 immune cells can be abolished e.g., by using antibodies against receptor this cytokine - anti-IL-10R. Furthermore, anticancer activity enhanced modifying them produce proinflammatory cytokines, IL-12, IL-15 or IL-18. Additionally, an immunomodulatory dose methotrexate hydroxyethyl starch (HES-MTX) nanoconjugate stimulate effector eliminate regulatory T which should enhance antitumor action immunotherapy DC vaccines. main aim our study was determine whether HES-MTX administered before with anti-IL-10R would change effect overproducing IL-15, Methods modified DCs checked two therapeutic protocols addition anti-IL10R chemoimmunotherapy antibodies. inhibition therapy inducing specific response were determined analyzing lymphoid myeloid cell populations nodules, restimulated splenocytes.
