This study investigated the presence, distribution, and antimicrobial resistance profiles of extended-spectrum beta-lactamase (ESBL)-producing Escherichia coli in a dairy herd located Northern Italy. The feces clinically healthy calves, their mothers, cows treated for mastitis, as well water, environmental samples, waste milk were collected subjected to bacteriological culture on CHROMagarTM ESBL plates. A questionnaire was administered identify risk factors. isolates identified E. by MALDI-TOF MS double-disk synergy test (DDST) minimal inhibitory concentration (MIC) assay. As result, isolated from 28 37 (75.67%) 2 3 (66.67%) cows, 8 14 (57.15%) milk. All showed multiple resistances categorized multidrug-resistant (MDR). Several factors selection diffusion identified, including lack routine cleaning calf feeding housing equipment, administration male blanket dry cow therapy. In conclusion, this highlighted presence MDR, most association with different sample sources. Accordingly, adding prudent use antibiotics, adoption adequate farm hygiene biosecurity measures might also help prevent spread transmission within herd.

Wastewater systems are usually considered antibiotic resistance hubs connecting human society and the natural environment. Antibiotic usage can increase abundance of both ARGs (antibiotic genes) MGEs (mobile gene elements). Understanding transcriptomic profiles remains a major research goal. Batch experiments were conducted by adding ampicillin to fresh activated sludge final concentrations 100 20 mg/L. The concentration was detected via UPLC‒MS/MS. Metatranscriptomic data from samples in reactors time series (0 10 h). transcription summarized searching metatranscriptomic datasets against related databases. During incubation, dosed could be removed completely within 8.5 h 4 h, following first-order biodegradation kinetics models (R2: 0.983–0.991). phylogenetic composition ARG at DNA level remained stable. At transcriptional level, significant upregulation class A beta-lactamase (P14171 gene) observed. P14171 is mostly spread wastewater treatment carried Rhodobacteraceae. Transcription (especially TnpA) sul1 stimulated external ampicillin. However, no correlations (ρ > 0.7, p < 0.01) between across all detected. only key that responded degradation tested systems, indicating its ability potential health risk.
