Oxidative stress plays a critical role in various physiological and pathological processes, particularly during pregnancy, where it can significantly affect maternal fetal health. In the context of viral infections, such as those caused by Human Immunodeficiency Virus (HIV) severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), oxidative may exacerbate complications disrupting cellular function immune responses. Antiviral drugs, while essential managing these also contribute to stress, potentially impacting both mother developing fetus. Understanding mechanisms which antivirals examination pharmacokinetic changes pregnancy that influence drug metabolism is essential. Some research indicates antiretroviral drugs induce mitochondrial dysfunction other studies suggest their use generally safe. Therefore, concerns about long-term health effects persist. This review delves into complex interplay between antioxidant defenses, antiviral therapies, focusing on strategies mitigate potential damage. By addressing gaps our understanding, we highlight importance balancing efficacy with risks stress. The liver is an active metabolic site that generates high levels of reactive oxygen species (ROS). Oxidative stress has been implicated in the chronicity hepatitis and B virus (HBV) infection. This study aimed to determine involvement oxidative HBV-infected cells efficacy natural Nrf2 activators. intracellular HBV pregenomic RNA copy number relative total was measured by RT-PCR, various protein expressions associated with were analyzed a Western blot analysis. results showed Nrf2, HO-1, Akt, Bcl-xL proteins decreased continuous infection, indicating HBV-positive exposed stress. present evaluated anti-HBV infection effects activator fucoxanthin (Fx), marine carotenoid from edible biological resources, including comparative pteryxin (Ptx). These activators suppressed production cells, thus increasing expression HO-1. In persistently infected transfected genome, Keap1 proteins, which contribute suppressing HBx involved replication, overexpressed. particular, activity these marked at low concentrations Fx. suggests may play significant role could be valuable source for further development through functional utilization food resources.

Background and Aims: Protein tyrosine sulfation (PTS) is a common posttranslational modification that regulates variety of physiological pathological processes. However, the role PTS in cancer remains poorly understood. The goal this study was to determine whether how plays HCC progression. Approach Results: By mass spectrometry bioinformatics analysis, we identified SAV1 as novel substrate HCC. Oxidative stress upregulates transcription SLC35B2, Golgi-resident transporter sulfate donor 3ʹ-phosphoadenosine 5ʹ-phosphosulfate, leading increased SAV1. Sulfation disrupts formation SAV1-MST1 complex, resulting decrease MST1 phosphorylation subsequent inactivation Hippo signaling. These molecular events ultimately foster growth cells both vivo vitro. Moreover, SLC35B2 target gene pathway, constituting positive feedback loop facilitates progression under oxidative stress. Conclusions: Our findings reveal regulatory mechanism SLC35B2/SAV1 axis response stress, highlighting its potential promising therapeutic for

Diffuse large B-cell lymphoma (DLBCL) is an aggressive characterized by distinct subtypes and heterogeneous treatment outcomes. Oxidative stress the dysregulation of related regulatory genes are prevalent in DLBCL, prompting investigation into nuclear factor erythroid 2-related 2 (Nrf2)-kelch-like ECH-associated protein 1 (Keap1) signaling pathway associated genes. The present study assessed pathological specimens clinical data from 43 newly diagnosed patients with comparing associations correlations between expression Nrf2, Keap1, microtubule-associated light chain 3β (LC3B) nitrotyrosine activated (ABC) germinal center (GCB) DLBCL using immunohistochemistry digital image analysis software. Nuclear Nrf2 activation was observed 33.3% ABC, demonstrating a higher prevalence hepatitis B surface antigen positivity, calcium ions significant body weight loss (P<0.05). Total GCB subtype inversely correlated Keap1 ABC subtype. Furthermore, positive correlation demonstrated LC3, indicating that total inhibited regulates LC3 expression. also lower white blood cell counts more frequent chemotherapy courses than These findings suggest could be biomarker for diagnosis.

Background and purpose: Diet might be a modifiable factor in preventing cancer by modulating inflammation. This study aims to explore the association between dietary inflammatory index (DII) score risk of bladder (BC). Methods: A total 112 BC patients 292 control subjects were enrolled case–control trial. Additionally, we tracked 109 319 controls, whose propensity scores obtained from Nutrition Examination Survey (NHANES) database spanning 1999 2020. The baseline intake data assessed using food frequency questionnaire (FFQ). DII calculated based on 20 nutrients participants categorized into four groups. potential diet was investigated multivariate odds ratios (ORs) 95% confidence intervals (CIs). Results: High associated with pro-inflammatory higher BC, positively (quartiles 4 vs. 1, ORs 4.89, CIs 2.09–11.25 p < 0.001). Specifically, this promote development inducing oxidative stress affecting DNA repair mechanisms. result consistent NHANES findings 2.69, 1.25–5.77, = 0.006) further supported lifestyle factors BC. Hepatocellular carcinoma (HCC) comprises the majority of primary liver cancers and possesses a low 5-year survival rate when in advanced stages. Anillin (ANLN), key player cell growth cytokinesis, is implicated HCC development. Currently, no treatment agents are known to suppress ANLN. Analysis The Cancer Genome Atlas data showed that high ANLN expression associated with poor prognosis patients. knockdown was shown inhibit proliferation, cycle progression, PD-L1 cancer cells. antipsychotic drug penfluridol identified Connectivity Map analysis. Penfluridol downregulated at both mRNA protein levels, leading G2/M arrest reduced colony formation Mechanistically, inhibited transcription factor MYC from binding an E-box motif promoter. This process mediated by penfluridol-induced upregulation NRF2, which competitively bound sequestered away interaction between NRF2 KEAP1, increasing NRF2. In syngeneic mouse model, suppressed tumour accompanied increased decreased expression. These findings suggest can be applied as first blocker modulate MYC/NRF2/KEAP1 axis.
