Prior research has proposed a potential association between lung cancer and inflammatory cytokines, yet the specific causal relationship remains unclear, especially across various pathologies. This study utilized bidirectional Mendelian randomization (MR) to explore these connections, unveiling novel insights. Our revealed distinctive cytokine profiles for each subtype of identified biomarkers that could refine diagnostic therapeutic approaches. We applied two-sample randomization, leveraging genetic variance data from three extensive genome-wide studies (GWAS) focusing on different types (lung adenocarcinoma: 1590 cases 314,193 controls healthy individuals European descent; squamous cell carcinoma: 1510 ancestry; small cancer: 717 ancestry). A separate GWAS summary cytokines 8,293 participants was also included. Dinaciclib, a potent cyclin-dependent kinase (CDK) inhibitor, has demonstrated considerable antitumor effects in various malignancies. However, its impact on oral squamous cell carcinoma (OSCC), predominant and highly aggressive form of head neck (HNSC) with limited treatment options, remains underexplored. We conducted gene set enrichment analyses HNSC patients that reinforced the relevance these cycle-related genes to OSCC pathogenesis. Given known dysregulation patients, we hypothesized Dinaciclib may inhibit growth by targeting overexpressed cyclins CDKs, thereby disrupting cycle progression inducing apoptosis. This study investigated Dinaciclib's proliferation, progression, apoptosis lines Ca9-22, OECM-1, HSC-3. Our results significantly reduces proliferation dose-dependent manner. Abstract Breast cancer is the most prevalent among women. Previous studies demonstrated that adipose-derived stem cells (ADSCs) co-cultured with resistin promote malignant behaviors in breast cells. This study explores roles of ADSCs and adipocytokine within tumor microenvironment, emphasizing their contributions to metabolic reprogramming progression. RNA sequencing analysis pathways revealed co-culture resistin-treated exhibited elevated expression NDUFS1, largest subunit mitochondrial complex I. Knockdown NDUFS1 inhibited cell proliferation tumorsphere formation, whereas its overexpression enhanced these effects through metabolism-mediated PROX1/c-Myc signaling pathway. Furthermore, treatment metformin, an inhibitor NDUFS1-activated metabolism, reduced Myc PROX1 diminished proliferation. Syngeneic orthotopic mouse model showed downregulation significantly mammary growth alongside decreased c-Myc. Also, metabolism inhibitors metformin rotenone a therapeutic effect on NDUFS1-expressing tumor. Collectively, this research establishes novel mechanistic framework linking adaptations cancer, paving way for innovative strategies aimed at targeting signaling.

Abstract This study aimed to investigate the effect and mechanism of 2α, 3α, 24‐thrihydroxyurs‐12‐en‐24‐ursolic acid (TEOA) alone or in combination with cisplatin on oral cancer. TEOA, a pentacyclic triterpenoid compound isolated from roots Actinidia eriantha , has demonstrated antitumor activity preclinical experiments. However, its role cancer remains poorly understood. Our findings revealed that low concentration TEOA did not exhibit significant cytotoxicity against squamous cell carcinoma cells. when combined cisplatin, showed therapeutic effect.
