Advanced machine learning (ML) and deep (DL) methods have recently been utilized in Drug Response Prediction (DRP), these models use the details from genomic profiles, such as extensive drug screening data cell line data, to predict response of drugs. Comparatively, DL-based prediction approaches provided better features. However, prior knowledge, like pathway is sometimes discarded irrelevant since datasets are multidimensional noisy. Optimized feature extraction processes suggested handle this problem. First, noise class imbalance problems must be tackled avoid low identification accuracy, long times, poor applicability. This article aims apply Non-Negativity-Constrained Auto Encoder (NNCAE) network tackle issues, enhance adaptive search for optimal size sliding windows, ensure that architectures adept at vital hidden NNCAE methodology used after performing standard pre-processing procedures balanced noise-removed input features learned train proposed hybrid classifier. The classification model, Golden Eagle Optimization-based Convolutional Long Short-Term Memory neural networks (GEO-Conv-LSTM), assembled by integrating Neural Network CNN LSTM models, with parameter tuning performed GEO algorithm. Evaluations conducted on two large Genomics Sensitivity Cancer (GDSC) repository, NNCAE-GEO-Conv-LSTM-based approach has achieved 96.99% 97.79% accuracies, respectively, reduced processing time error rate DRP

Obesity has emerged as an important global health challenge, significantly influencing the incidence and progression of various cancers. This comprehensive review elucidates complex relationship between obesity oncogenesis, focusing particularly on role dysregulated signaling pathways central mediators this association. We delve into contributions obesity-induced alterations in key cascades, including PI3K/AKT/mTOR, JAK/STAT, NF-κB, Wnt/β-catenin to carcinogenesis. These facilitate unchecked cellular proliferation, chronic inflammation apoptosis resistance. Epidemiological evidence links with increased cancer susceptibility adverse prognostic outcomes, pronounced risks for specific cancers such breast, colorectal, endometrial hepatic malignancies. synthesizes data from both animal clinical studies underscore pivotal disrupted shaping innovative therapeutic strategies. highlight critical importance lifestyle modifications management risk mitigation, stressing benefits dietary changes, physical activity, behavioral interventions. Moreover, we examine targeted pharmacological strategies addressing aberrant obesity-related tumors discuss integration cutting-edge treatments, immunotherapy precision medicine, practice.

Abstract Background Breast cancer manifests as a heterogeneous pathology marked by complex metabolic reprogramming essential to satisfy its energy demands. Oncogenic signals boost the metabolism, modifying fatty acid synthesis and glucose use from onset progression therapy resistant-forms. However, exact contribution of dependencies during tumor evolution remains unclear. Methods In this study, we elucidate connection between FASN LDHA, pivotal genes, their correlation with grade response using datasets public repositories. Subsequently, evaluated proliferative functions upon LDHA inhibition in breast models. Lastly, integrated metabolomic lipidomic analysis define contributions metabolites, lipids, precursors phenotypes. Results Collectively, our findings indicate shifts progression, unvealling two distinct functional phenotypes associated aggressiveness response. Specifically, exhibits reduced expression advance-grade tumors therapy-resistant forms, whereas demonstrates higher expression. Additionally, biological impact blocking enzymatic activity was correlated resistant conditions. Conclusions These observations emphasize intrinsic heterogeneity within cancer, thereby highlighting relevance interventions field precision medicine.

Background: The aim of this study was to determine the effect Sparstolonin B (SsnB) on cell proliferation and apoptosis in human breast cancer (MCF-7) ovarian epithelial (OVCAR-3) lines presence absence estradiol hemihydrate (ES). Phosphoinositol-3 kinase (PI3K), phosphorylated protein alpha (p-AKT), mTOR (mechanistic target rapamycin) signaling proteins, sphingomyelin/ceramide metabolites were also measured within scope study. Methods: anti-proliferative effects SsnB therapy evaluated over a range times concentrations. Cell determined by measuring Proliferating Nuclear Antigen (PCNA). PCNA quantified ELISA distribution assessed immunofluorescence microscopy. MTT analysis used test vitality cells, while LC-MS/MS analyze amounts ceramides (CERs), sphingosine-1-phosphate (S1P), sphingomyelins (SMs). TUNEL labeling assess apoptosis, staining enzyme-linked immunosorbent assay (ELISA) measure levels PI3K, p-AKT, p-mTOR proteins. Results: administration significantly decreased viability MCF-7 OVCAR-3 cells both ES, it did not cause toxicity healthy fibroblasts. In comparison controls, treated with showed significant drop S1P, p-mTOR. cultured SsnB, increase intracellular concentrations C16-C24 CERs observed. Conclusions: downregulated reducing promoting ceramide buildup apoptosis.
