Summary The abundance of various cell types can vary significantly among patients with varying phenotypes and even those the same phenotype. Recent scientific advancements provide mounting evidence that other clinical variables, such as age, gender, lifestyle habits, also influence certain types. However, current methods for integrating single-cell-level omics data variables are inadequate. In this study, we propose a regularized Bayesian Dirichlet-multinomial regression framework to investigate relationship between single-cell RNA sequencing patient-level data. Additionally, model employs novel hierarchical tree structure identify relationships at different cell-type levels. Our successfully uncovers significant associations specific across three distinct diseases: pulmonary fibrosis, COVID-19, non-small lung cancer. This integrative analysis provides biological insights could potentially inform interventions diseases.

Background One of the effective strategies for treatment tumor diseases, including colitis-associated colorectal cancer (CAC), is immunotherapy. During inflammation, NF-κB activated, which connected with hypoxia-inducible factor—HIF, regulating immune cells functioning and influences CAC development. Organisms differ according to their hypoxia resistance HIF expression. Therefore, aim study was characterize thymus, spleen mesenteric lymph nodes morphofunctional features, as well changes in subpopulation composition peripheral blood tolerant susceptible C57Bl/6 mice CAC. Methods Hypoxia tolerance assessed by gasping time measurement hypobaric decompression chamber. Based on outcome, were assigned three groups characterized ‘tolerant hypoxia’, ‘normal’, ‘susceptible hypoxia’. A month after determining modeled intraperitoneal azoxymethane (AOM) administration cycles dextran sulfate sodium consumption. Mice sacrificed 141st day AOM administration, a morphological, morphometric immunohistochemical tumors, morphological thymus spleen, assessment carried out. Results Tumors represented glandular intraepithelial neoplasia adenocarcinomas, area larger mice. Immunohistochemical revealed more pronounced Ki-67+ staining tumors In CAC, only mice, expansion thymic cortex observed relative control group, while ones, no detected. Only germinal centers lymphoid follicles enlargement observed. during comparison absolute number B-lymphocytes relative—cytotoxic T-lymphocytes increased. The cytotoxic NK higher compared ones. revealed—only them cells, increased, macrophages decreased. Conclusions Morphofunctional differences reactions indicated response development should be taken into account experimental studies.
