To study the composition and succession of bacterial fungal communities during fermentation cigar filler leaves with varying initial water contents, high-throughput sequencing technology was used to sequence 16SrRNA genes ITS1 from tobacco leaf samples. This followed by analyses microbial α-diversity, community structure, function prediction based on data. The diversity richness decreased over time under different content conditions. Among 18 samples, predominant phyla identified were Proteobacteria, Firmicutes, Actinobacteria, Ascomycota, Basidiomycota, leading genera being Staphylococcus, Sphingomonas, Methylobacterium-Methylorubrum, Pseudomonas, Humicola. Functional predictions for bacteria revealed their primary involvement in carbohydrate, lipid, amino acid metabolism. influenced structure relative abundance fermentation. While exhibited a similar structural composition, there notable differences abundance. functional results PICRUSt indicated that predicted species among samples minimal, whereas variations more pronounced across stages contents.

ABSTRACT Rationale The cigars have characteristic flavors in smoke when compared with cigarettes, and from various origins also a difference taste. However, little information can be found about the chemical components of smokes between cigar cigarette as well cigars, so it is interesting to compare their pyrolysis product distribution. Methods leaves were pyrolyzed microfluidized‐bed reactor, vapors condensed collected using cold traps. Mass spectrometric analysis liquids was performed utilizing electrospray ionization‐orbitrap mass spectrometry both positive negative ion modes. Results spectra condensable products three tobacco obtained by Orbitrap‐MS ESI DBE values (to carbon atom number) relative distribution nitrogen or oxygen different carefully discussed. Conclusion main relates N 1 –N 2 class compounds high‐mass range highly unsaturated nitrogenated compounds, while one two associated C x H y O z 5–9 30‐50 w compounds. Besides, oxygenated that fall into H/C > O/C < ranges are for Dominican cigar.

Introduction In order to enhance the quality of cigar tobacco leaves (CTLs), a gradient variable temperature fermentation approach was employed. Methods The demonstrated gradual increase from low (35 ± 2°C) moderate (45 2°C), and then high (55 2°C). Each underwent 10-day process, resulting in total duration 30 days. Changes sensory evaluation, chemical composition, bacterial absolute quantitative structure function were examined throughout process fermentation. Results Compared constant fermentation, improved CTLs, reduced sugar cembrane degradation products, increased amino acid contents. It resulted significant changes quantity but had no effects on richness diversity communities. results correlation analyses showed that with which effected by predominant microbes. three-phase model characterize alterations CTLs. Conclusion positive influence CTL providing optimal for growth, generation quality-related compositions.
