Abstract Osteoporosis is characterized by an imbalance between osteoblasts and osteoclasts coupling excessive oxidative stress in the bone microenvironment that impairs defect healing increases risk of non‐union. In this study, injectable gypsogenin (GN)‐based organic–inorganic composite hydrogel (CCT/nHA@GN) developed to treat osteoporotic defects. The made grafting sodium citrate (SC) nano‐hydroxyapatite (nHA)/GN nanoparticles onto carboxymethylated chitosan (CMCS). GN a natural small‐molecule saponin, which shows biocompatibility anti‐oxidant properties. resulting well‐defined porous structure, favorable degradability, controlled drug‐release properties, suitable rheological characteristics. Importantly, it reverses differentiation fate marrow‐derived mesenchymal stem cells (BMSCs) from patients promotes angiogenesis human umbilical vein endothelial (HUVECs). Furthermore, activates AMPKα‐FOXO3a‐CAT/MnSOD signaling pathway via AMPKα phosphorylation, thereby augmenting antioxidant capacity, promoting osteogenesis, inhibiting osteoclastogenesis, ultimately rectifying disrupted microenvironment. vivo studies reveal volume total (BV/TV) ratio bones regenerated with CCT/nHA@GN 2.85 times higher than control group. conclusion, these findings suggest can be promising alternative material for treatment
