Cardiovascular
disease
comprises
a
group
of
disorders
affecting
or
originating
within
tissues
and
organs
the
cardiovascular
system;
most,
if
not
all,
will
eventually
result
in
cardiomyocyte
dysfunction
death,
negatively
impacting
cardiac
function.
Effective
models
are
thus
important
for
understanding
crucial
aspects
progression,
while
recent
advancements
stem
cell
biology
have
allowed
use
populations
to
derive
such
models.
These
include
three-dimensional
(3D)
as
cell-based
embryos
(SCME)
well
organoids,
many
which
frequently
derived
from
embryoid
bodies
(EB).
Not
only
can
they
recapitulate
3D
form
function,
but
developmental
programs
governing
self-organization
into
more
complex
well.
Many
different
organoids
SCME
constructs
been
generated
years
recreate
tissue
that
give
rise
its
cellular
composition
unique
morphology.
It
is
purpose
this
narrative
literature
review
describe
summarize
recently
organoid
their
recapitulation
genetic
acquired
disease.
Owing
examined,

injury

with

Early
embryonic
development
is
a
complex
process
where
undifferentiated
cells
lose
their
pluripotency
and
start
to
gastrulate.
During
gastrulation,
three
germ
layers
form,
giving
rise
different
cell
lineages
organs.
This
regulated
by
transcription
factors
epigenetic
regulators,
including
non-canonical
polycomb
repressive
1s
(ncPRC1s).
Previously,
we
reported
that
ncPRC1-member
RYBP
(RING1
YY1
binding
protein)
crucial
for
implantation
cardiac
lineage
commitment
in
mice.

Abstract
Mammalian
specification
of
mesoderm
and
definitive
endoderm
(DE)
is
instructed
by
the
two
related
Tbx
transcription
factors
(TFs)
Eomesodermin
(
Eomes
)
Brachyury
sharing
partially
redundant
functions.
Gross
differences
mutant
embryonic
phenotypes
suggest
specific
functions
each
TF.
To
date,
molecular
details
separated
lineage-specific
gene-regulation
remain
poorly
understood.
Here,
we
combine
stem
cell-based
analyses
to
delineate
non-overlapping,
transcriptional
activities.
On
a
genome-wide
scale
binding
both
TFs
overlaps
at
promoters
target
genes,
but
shows
specificity
for
distal
enhancer
regions,
that
conferred
in
DNA-binding
motifs.
The
unique
sites
instructs
anterior
(AM)
DE
caudal
.
Remarkably,
EOMES
antagonizes
BRACHYURY
gene-regulatory
co-expressing
cells
during
early
gastrulation

proper
sequence

Mammalian
embryos
are
very
vulnerable
to
environmental
toxicants
(ETs)
exposure.
Bisphenol
A
(BPA),
one
of
the
most
diffused
ETs,
exerts
endocrine-disrupting
effects
through
estro-gen-mimicking
and
hormone-like
properties,
with
detrimental
health
effects,
including
on
reproduction.
However,
its
impact
during
peri-implantation
stages
is
still
unclear.
This
study,
using
gastruloids
as
a
3D
stem
cell-based
in
vitro
model
embryonic
development,
showed
that
BPA
exposure
arrests
their
axial
elongation
when
present
Wnt/β-catenin
pathway
activation
period
by
β-catenin
protein
reduction.
Gastruloid
reshaping
might
have
been
impeded
downregulation
Snail,
Slug
Twist,
known
suppress
E-cadherin
expression
activate
N-cadherin
gene,
low
protein.
Also,
lack

BPA-exposed
