How chromatin bridges are detected by the abscission checkpoint during mammalian cell division is unknown. Here, we discuss recent findings from our lab showing that DNA topoisomerase IIα (Top2α) enzyme binds to catenated ("knotted") next midbody and forms abortive Top2-DNA cleavage complexes (Top2ccs) on bridges. Top2ccs then processed proteasome promote localization of damage sensor protein Rad17 Top2-generated double-strand ends knots. In turn, promotes local recruitment MRN complex downstream ATM-Chk2-INCENP signaling delay prevent bridge breakage in cytokinesis.
