Distinct UPR and Autophagic Functions Define Cell-Specific Responses to Proteotoxic Stress in Microglial and Neuronal Cell Lines

Autophagy
is
a
catabolic
process
involved
in
different
cellular
functions.
However,
the
molecular
pathways
governing
its
potential
roles
cell
types
remain
poorly
understood.
We
investigated
role
of
autophagy
context
proteotoxic
stress
two
central
nervous
system
types:
microglia-like
line
BV2
and
neuronal-like
N2a.
Proteotoxic
stress,
induced
by
proteasome
inhibition,
produced
early
apoptosis
cells,
due
part
to
predominant
activation
PERK-CHOP
pathway.
In
contrast,
N2a
cells
showcased
greater
resistance
robust
induction
IRE1α-sXbp1
arm
UPR.
also
demonstrated
that
activated
both
lines
but
with
kinetics
restored
proteostasis,
while
it
participated
regulating
phagocytosis.
Finally,
predominantly
mTORC2-AKT-FOXO1-β-catenin
pathway
preferentially
PDK1-AKT-FOXO3
axis.
Collectively,
our
findings
suggest
triggers
cell-specific
responses
microglia
neurons,
physiological
outcomes.
