Bactrocera dorsalis Hendel (Diptera: Tephritidae) is a globally invasive pest that inflicts severe economic damage on various fruit and vegetable species.Deltamethrin commonly employed as an insecticide against B. dorsalis. This study explored the intergenerational sublethal effects of deltamethrin dorsalis.Sublethal low lethal exposure to reduced mating rates by inhibiting production sex pheromones (2,3,5-trimethylpyrazine 2,3,5,6-tetramethylpyrazine) in male adult parental (F 0 ) generation.Due down-regulation sarcosine dehydrogenase expression, essential for pheromone biosynthesis.Furthermore, three tested concentrations (LC 5 , LC 20 50 hatching rate, pupation emergence rates, reproduction several population parameters progeny 1 generation, including intrinsic rate increase (r); finite (λ); net reproductive (R gross (GRR) following F exposure.Additionally, insect hormone levels (Juvenile Hormone 20-hydroxyecdysone) showed significant changes female adults treated with deltamethrin.Furthermore, genes related reproduction, such Vg1, Vg2, Vg3, VgR, exhibited both generations.Importantly, fecundity decreased generation 5, deltamethrin, although increased females at concentrations. This highlights intricate impacts dorsalis, revealing generations.

MiRNAs are critical regulators of numerous physiological and pathological processes. Ascosphaera apis exclusively infects bee larvae causes chalkbrood disease. However, the function mechanism miRNAs in larval response to A. infection is poorly understood. Here, ame-miR-34, a previously predicted miRNA involved Apis mellifera invasion, was subjected molecular validation, overexpression knockdown were then conducted explore regulatory functions ame-miR-34 body weight immune response. Stem-loop RT-PCR Sanger sequencing confirmed authenticity gut mellifera. RT-qPCR results demonstrated that compared with uninfected guts, expression level significantly downregulated (p < 0.001) guts apis-infected 4-, 5-, 6-day-old larvae, indicative remarkable suppression host due infection. In comparison corresponding negative control (NC) groups, mimic-miR-34 group upregulated 0.001), while inhibitor-miR-34 0.01). Similarly, effective achieved. addition, weights 5- increased those mimic-NC group; 5-day-old decreased inhibitor-NC group, 4- increased, indicating involvement modulating weight. Furthermore, levels both hsp abct after overexpression. contrast, knockdown, aforementioned two key genes downregulated. Together, noninfected could be achieved by feeding method, exerted invasion through positive regulation abct. Our findings not only provide valuable reference for functional investigation but also reveal role Additionally, this study may promising target treatment

MicroRNAs (miRNAs) are small single-stranded non-coding RNAs involved in a variety of cellular events by regulating gene expression at the post-transcriptional level. Several core genes miRNA biogenesis have been reported to participate wide range physiological events, some insect species. However, functional significance pathway Nilaparvata lugens remains unknown. In present study, we conducted systematic characterisation five biogenesis. We first performed spatiotemporal analysis and found that exhibited similar patterns, with high levels eggs relatively transcriptional ovaries fat bodies females. RNA interference experiments showed injecting third-instar nymphs dsRNAs targeting genes, NlAgo1, NlDicer1, NlDrosha resulted mortality rates various degrees body melanism, moulting defects, wing deformities. Further investigations revealed suppression severely impaired ovarian development oocyte maturation, resulting significantly reduced fecundity disruption intercellular spaces between follicle cells. Moreover, profiles miR-34-5p, miR-275-3p, miR-317-3p, miR-14, Let-7-1, miR-2a-3p were altered response knockdown mixture, suggesting they play essential roles miRNA-mediated expression. Therefore, our results provide solid theoretical basis for N. suggest NlDrosha-dependent reproduction this agricultural pest.

Abstract BACKGROUND The oriental fruit fly, Bactrocera dorsalis , causes significant economic losses in production because of its high reproductive rate and adaptability. current study investigated the sublethal lethal effects avermectin, a macrocyclic lactone insecticide, on F 0 1 generations B. . RESULTS This demonstrated that exposure to avermectin with (LC 5 LC 20 50 ) concentrations reduced mating rates, prolonged pre‐mating periods, lowered levels juvenile hormone (JH) 20‐hydroxyecdysone (20E). Furthermore, resulted decrease vitellogenin ( Vg receptor VgR levels, downregulated expression corresponding genes. Subsequent investigations revealed led sugar accumulation indicated by decreased glycogen, glucose trehalose alongside impaired trehalase activities lower triglyceride content generation These metabolic disruptions contributed hatching, pupation emergence pupal weights parameters (as life table parameters) generation. increased detoxifying enzymes such as glutathione S ‐transferase, carboxylesterase cytochrome P450 monooxygenases were found, suggesting an adaptive response detoxify avermectin. CONCLUTION findings highlight potential long‐term impacts population dynamics underscore need for incorporating considerations into integrated pest management strategies. © 2025 Society Chemical Industry.

MiRNAs, as a kind of key regulators in gene expression, play vital roles numerous life activities from cellular proliferation and differentiation to development immunity. However, little is known about the regulatory manner miRNAs Asian honey bee (Apis cerana) guts. Here, on basis our previously gained high-quality transcriptome data, transcriptome-wide identification larval guts Apis cerana was conducted, followed by investigation miRNAs' differential expression profile during gut development. In addition network, potential function differentially expressed (DEmiRNAs) further analyzed. total, 330, 351, 321 were identified 4-, 5-, 6-day-old guts, respectively; among these, 257 shared, while 38, 51, 36 ones specifically expressed. Sequences six confirmed stem-loop RT-PCR Sanger sequencing. Additionally, "Ac4 vs. Ac5" comparison group, there seven up-regulated eight down-regulated miRNAs; these DEmiRNAs could target 5041 mRNAs, involving series GO terms KEGG pathways associated with growth development, such process, cell part, Wnt, Hippo. Comparatively, four detected "Ac5 Ac6" group targets diverse development-related pathways, including cell, organelle, Notch Wnt. Intriguingly, it noticed that miR-6001-y presented continuous up-regulation trend across developmental process implying may be essential modulator Further indicated 43 31 engaged several crucial development-associated signaling Hippo, Notch. Ultimately, trends five randomly selected verified using RT-qPCR. These results demonstrated dynamic structural alteration accompanied A. c. likely participate modulation well affecting critical via regulation genes. The RNA interference pathway mediated by microRNAs (miRNAs) is one of the methods to defend against viruses in insects. Recent studies showed that miRNAs participate viral infection binding target genes regulate their expression. Here, we found Bombyx mori miRNA, miR-6498-5p was down-regulated, whereas its predicted gene pyridoxal phosphate phosphatase PHOSPHO2 (BmPLPP2) up-regulated upon nucleopolyhedrovirus (BmNPV) infection. Both vivo and vitro experiments targets BmPLPP2 suppresses Furthermore, inhibits BmNPV genomic DNA (gDNA) replication, promotes gDNA replication. As a (PLP) (PLPP), overexpression results reduction PLP content, knockdown leads an increase content. In addition, exogenous replication gDNA; contrast, inhibitor 4-deoxypyridoxine facilitates Taken together, concluded has potential anti-BmNPV role down-regulating modulate but induces down-regulation promote proliferation. Our findings provide valuable insights into host miRNA B. mori-BmNPV interaction. identification antiviral molecule offers novel perspective on strategies for preventing managing sericulture.
