Abstract Heterozygosity is a measure of allelic diversity within individuals. Puccinia striiformis f. sp. tritici ( Pst ) highly heterozygous dikaryotic crop pathogen. Its source heterozygosity variation and the contribution to adaptability are still unknown. By analyzing resequencing data 266 worldwide isolates, we found that may have undergone an important historical hybridization event, introducing substantial leading divergence its haploid genomes into two distinct haplotypes. Strains with both haplotypes exhibit higher individual allele wider geographical distribution. Between genomes, 16% genome had diverged, scattered as mosaic blocks genome. These regions enriched genes displaying critical roles during infection plant host, expression levels. In these regions, 8.0 Mb shows recombination fingerprints associated virulence, while 4.9 displays linkage across entire We demonstrated sexual in extensive significant. can gain genetic due intra- inter-species hybridization. Permanent heterozygous loci, such as sex- or mating-compatibility regions, often display suppression of recombination and signals genomic degeneration. In Basidiomycota, two distinct loci confer mating compatibility. These encode homeodomain ( HD ) transcription factors pheromone receptor Pra )-ligand allele pairs. To date, an analysis genome level mating-type (MAT) is lacking for obligate biotrophic basidiomycetes in the Pucciniales , order containing serious agricultural plant pathogens. Here, we focus on four species Puccinia that infect oat wheat, including P . coronata f. sp. avenae graminis tritici triticina striiformis MAT are located separate chromosomes supporting previous hypotheses a tetrapolar compatibility system The genes multiallelic all while PR locus appears biallelic, except which potentially has multiple alleles. largely conserved their macrosynteny, both within between species, without strong suppression. Abstract Basidiomycetes like rust fungi have complex reproductive cycles and dikaryotic life stages which influence their population structure evolution. Puccinia coronata f. sp. avenae ( Pca ), the causal agent of oat crown rust, is a pathogen global economic importance. To investigate genetic diversity species, as well role mating type system nuclear exchange (somatic hybridization) in host adaptation we acquired whole genome sequencing data Taiwanese Australian isolates, adding to existing for USA South African populations. An atlas 30 chromosome-level, fully-phased haplotypes from six isolates nine was generated capture genomic composition key lineages. This study provides evidence recombination both populations mechanisms introduction diversity. The limitations assuming clonal evolutionary history virulence phenotyping demonstrated by detection either sexual or cryptic population. Overall, characterization intercontinental migration at haplotype level further impetus molecular monitoring on scale.

Cereal rusts can reproduce sexually or asexually, and emergence novel lineages has potential serious epidemics one caused by 'Warrior' lineage in Europe. In global context, Pst Canada were not well-characterized origin foreign incursions was known. Additionally, while some mating type genes have been identified published genomes, there no rigorous assessment diversity distribution across species.

In the absence of recombination, number transposable elements (TEs) increases due to less efficient selection, but dynamics such TE accumulations are not well characterized. Leveraging a dataset 21 independent events recombination cessation different ages in mating-type chromosomes Microbotryum fungi, we show that TEs rapidly accumulated regions lacking content reached plateau at ca. 50% occupied base pairs by 1.5 million years following suppression. The same superfamilies have expanded independently evolved non-recombining regions, particular rolling-circle replication (Helitrons). Long-terminal repeat (LTR) retrotransposons Copia and Ty3 also expanded, through transposition bursts (distinguished from gene conversion based on LTR divergence), with both autosomes affected, suggesting constitute reservoirs. This study improves our knowledge genome evolution showing can accumulate bursts, non-linear decelerating dynamics.

The population structure and evolution of basidiomycetes like rust fungi are influenced by complex reproductive cycles dikaryotic life stages where two independent nuclear haplotypes present in the cell. ability to alternate between asexual (clonal) sexual reproduction increases evolutionary capacity these species. Furthermore, exchange intact nuclei (somatic hybridization) can allow for rapid generation genetic variability outside cycle. Puccinia coronata f. sp. avenae (Pca), causal agent oat crown rust, is a pathogen global economic importance that difficult control due breakdown host resistance. contribution sexuality, clonality, migration virulence varies across Pca populations. As such, pathosystem ideal address role mating type, recombination, mutation, somatic hybridization adaptation. At haplotype level, we confirmed previous reports recombination additionally detected either or cryptic isolates, contrasting evaluations suggested Australia be purely clonal. also identified events not only associated with significant changes fitness but imply intercontinental haplotypes, which provides further impetus molecular monitoring on scale.

This paper discusses the mechanisms by which fungi manipulate plant physiology and suppress defense responses producing effectors that can target various host proteins. Effector-triggered immunity effector-triggered susceptibility are pivotal elements in complex molecular dialogue underlying plant-pathogen interactions. Pathogen-produced effector molecules possess ability to mimic pathogen-associated patterns or hinder binding of pattern recognition receptors. Effectors directly nucleotide-binding domain, leucine-rich repeat receptors, downstream signaling components defense. Interactions between these receptor-like kinases plants critical this process. Biotrophic adeptly exploit networks key hormones, including salicylic acid, jasmonic abscisic ethylene, establish a compatible interaction with their hosts. Overall, highlights importance understanding interplay fungal develop effective strategies for disease management.
