There is a lack of established biological, psychological, social, and digital markers for the prediction, identification, stratification patients with major depressive disorder (MDD). We therefore aimed to evaluate serum nerve growth factor (NGF) in MDD patients.In this case-control study, we recruited age- sex-matched healthy controls (HCs). A qualified psychiatrist evaluated study participants according Diagnostic Statistical Manual Mental Disorders, fifth edition. Serum NGF levels were analyzed using commercially available enzyme-linked immunosorbent assay kit.We data from 106 88 HCs. Mean concentrations significantly higher (104.70 ± 6.43 pg/mL) than HCs (72.09 7.69 pg/mL). Receiver operating characteristic curve analysis revealed good diagnostic performance MDD.Higher might be involved pathophysiology, altered may an early warning sign depression. Abstract Recently, scientists have focused on pro-inflammatory cytokines and immunological dysregulation in major depressive disorder (MDD). Some research suggests cytokines' role MDD development, whereas anti-inflammatory studies are sparse. There is no systematic investigation of Bangladeshi patients' pro- cytokines. This study examines the blood levels IL-12 IL-4 patients healthy controls (HCs) to determine diagnostic accuracy these identify from those without MDD. A total 110 people with department psychiatry a teaching hospital Dhaka 107 HCs participated this case–control study. Depression illness severity were gauged using DSM-5 criteria Ham-D scores. Commercially marketed ELISA kits used accordance manufacturer guidelines measure peripheral blood, allowing comparison patient control groups. In HCs, (5333.00 ± 307.40 pg/ml) showed noticeably higher than (2331.00 207.40 pg/ml). The increased positively correlated scores (male: r = 0.351, p < 0.050; female: 0.389, 0.050), suggesting possible relationship disease progression. Additionally, compared (272.81 23.94 pg/ml), had significantly (876.35 66.73 (p 0.001). Also, there was positive correlation between serum 0.361, 0.398, 0.050). Major Depressive Disorder (MDD) is a debilitating mental health condition with complex etiology, and recent research has focused on pro-inflammatory cytokines chemokines as potential contributors to its pathogenesis. However, studies investigating the roles of TNF-α MCP-4 in MDD within Bangladeshi population are scarce. This study aimed assess association between serum levels severity MDD, exploring their risk indicators for development.This case-control enrolled 58 patients from Bangabandhu Sheikh Mujib Medical University (BSMMU) Hospital, Dhaka, Bangladesh, alongside 30 age, sex, BMI-matched healthy controls. diagnosis followed DSM-5 criteria disease using 17-item Hamilton Depression Rating Scale (Ham-D). We measured ELISA assays according supplied protocols.The revealed significantly elevated (47±6.6 pg/ml, mean±SEM) compared controls (28.06±1.07 pg/ml). These increased positively correlated Ham-D scores (Pearson's r = 0.300, p 0.038), suggesting peripheral pathology. Additionally, exhibited higher (70.49±6.45 pg/ml) than (40.21±4.08 showed significant negative correlation (r -0.270, P 0.048) patients, indicating more role pathogenesis.This highlights that exhibit heightened inflammatory immune responses controls, supporting cytokine hypothesis Serum TNF-α, but not MCP-4, shows promise biomarker assessing development, which could aid early detection. Background Epidemiological research suggests that altered levels of cytokine are associated with pathophysiology and the development major depressive disorder (MDD). Based on earlier study, IL-1β rs16944 rs1143627 polymorphisms may increase risk depression. Here, we aimed to evaluate correlation between these MDD susceptibility among population in Bangladesh. Methods Blood samples were collected from 100 patients 70 matched controls. Study participants evaluated by DSM-5 criteria PCR-RFLP method applied for genotyping.
