This study aimed to investigate the therapeutic potential of Citrullus mucosospermus extract (CME) in counteracting adipogenesis and its associated metabolic disturbances murine models. In vitro experiments utilizing 3T3-L1 preadipocytes revealed that CME potently inhibited adipocyte differentiation, as evidenced by a dose-dependent reduction lipid droplet formation. Remarkably, also attenuated glucose uptake intracellular triglyceride accumulation fully differentiated adipocytes, suggesting ability modulate pathways mature adipose cells. Translating these findings an vivo setting, we evaluated effects C57BL/6N mice fed high-fat diet (HFD) for 10 weeks. administration, concomitantly with HFD, resulted significant attenuation body weight gain compared HFD control group. Furthermore, treatment led substantial reductions liver weight, total fat mass, deposits visceral retroperitoneal tissue, underscoring targeted impact on expansion. Histological analyses remarkable hepatic steatosis. While group exhibited severe within lobules, dose-dependently mitigated this pathology, highest dose virtually abolishing deposition. An examination tissue progressive hypertrophy upon treatment, culminating near-normalization morphology at dose. Notably, potent anti-inflammatory properties, significantly attenuating upregulation pro-inflammatory cytokines’ mRNA levels (TNF-α, IL-1β IL-6) livers HFD-fed mice. suggests mechanism through which may exert protective against inflammation obesity fatty disease.

Assessing the effect of various forms exercise training on patients with sarcopenic obesity. Two independent reviewers systematically searched English and Chinese databases (PubMed, Embase, Cochrane Library, Web Science, CNKI) for randomized controlled trials effects in obesity until October 2023. Reference materials grey literature were also included. Selected studies underwent screening, data extraction, quality assessment. Meta-analysis was conducted using Review Manager 5.4 software. A total 8 included final analysis. The results indicated that resistance (RT) significantly improved grip strength (MD = 3.85, 95%CI: 1.50 to 6.20, P < 0.01), percentage body fat -2.96, -4.19 -1.74, walking speed 0.23, 0.01 0.46, 0.04), IGF-1(MD 0.79, 0.05 1.52, 0.04) knee extension 4.85, 1.97 7.72, 0.01). There no statistically significant difference observed weight -0.61, -4.06 2.84, 0.73). Aerobic (AT) resulted a reduction among SO -6.07, -9.89 -2.25, while differences other outcome measures. Mixed (MT) -2.42, -3.58 -1.26, -4.40, -8.40 -0.40, 0.03), IGF-1 1.01, 0.45 1.56, 0.15, 0.04 0.26, However, -0.70, -4.00 2.60, 0.68) 1.73, -1.31 4.78, 0.26). RT, AT, MT could not improve level serum IL-6 SO, [MD -0.01,95%CI:-0.27 0.24, 0.92]. Various methods have differing muscle-reducing treatment. Compared aerobic training, mixed may offer more pronounced improvements, enhancing physical functioning patients. This underscores clinical significance intervention treating obesity, suggesting future explore intervention's role mechanism, particularly related IGF-1, IL-6, cytokines.

Sarcopenic obesity (SO) is a condition characterized by the coexistence of age-related and sarcopenia. This systematic review network meta-analysis (NMA) aimed to compare effects different training modalities, such as aerobic (AT), resistance (RT), combined with AT (CT), multicomponent (MCT) on body composition, muscle strength, physical performance in elderly patients SO. We electronically searched randomized controlled trials, published from inception March 2024 PubMed, Embase, Cochrane Library, Web Science Scopus. Effect estimates were presented mean differences (MD) or Standard Mean Difference (SMD) 95% confidence interval (CI). The comprehensive all treatments ranked surface under cumulative ranking (SUCRA) probabilities. 14 trials enrolling 955 participants included. fat percentage (BFP) MCT (MD= -6.37, CI: -8.67, -4.07), CT (MD = -2.08, -4.00, -0.16), RT -1.85, -3.25, -0.44) was significantly lower than normal control group, showing superior compared RT. Furthermore, only improved fat-free mass (FFM; MD 5.21, CI:1.51, 8.91), well index (BMI; 0.74, CI:0.08, 1.40). In addition, handgrip strength (HGS) both (SMD 0.87, 0.19, 1.5) RT(SMD 0.84, 0.43, 1.25). 30s chair stand test also yielded better outcomes for 3.10, 1.33, 4 0.86), CT(MD 2 0.50, 0.18, 5.18), RT(MD 3.91, 2.30, 5.52) when group. Lastly, gait speed enhanced 0.35, 0.30, 0.41) 0.14, 0.06, 0.21). results indicated that other modalities enhancing composition speed. contrast, appears be more advantageous 30-second standing improving HGS. outperformed beneficial These findings provide valuable insights clinicians researchers customize exercise prescriptions older http://www.crd.york.ac.uk/PROSPERO/display_record.asp?ID=CRD42024544962.

Background/Objective: Sarcopenic obesity (SO), a pathological syndrome characterized by the co-existence of diminished muscle mass and excessive adipose accumulation, significantly compromises quality life in older adults. The purpose this study was to systematically evaluate efficacy exercise, nutritional interventions, neuromuscular electrical stimulation (NMES) preventing managing SO middle-aged Methods: A comprehensive search conducted across PubMed, Web Science, Embase, Cochrane Library, CNKI for randomized controlled trials (RCTs) until January 2025. Meta-analyses were performed using random-effects model fixed-effects based on degree heterogeneity calculating mean differences (MD) with 95% confidence intervals (CI). Subgroup analyses compared intervention types. Results: Twenty-nine RCTs (1622 participants) included. Exercise interventions reduced body fat percentage (MD = −2.79%, CI: −3.94, −1.64, p < 0.001, I2 74%), −6.77 kg, −11.48, −2.06, 0.005, 98%), waist circumference −2.05 cm, −3.64, −0.46, 0.01, 0%) LDL-C (MD: −7.45 mg/dL, −13.82, −1.07, 0.02, 0%), while improving handgrip strength 2.35 1.99, 2.70, 52%) gait speed 0.19 m/s, 0.13, 0.24, 89%). Mixed training outperformed resistance-only regimens reducing enhancing functional outcomes. NMES −2.01%, −3.54, −0.48, 93%) −1.72 −2.35, −1.09, increasing Skeletal Muscle Index 0.26 kg/m2, 0.22, 0.29, 38%). Synergy supplementation amplified these effects. Nutritional modestly improved total fat-free 0.77 0.04, 1.50, 1.35 0.71, 2.00, but showed no significant impact metabolic markers (TG, TC, glucose, hemoglobin, HOMA-IR). Conclusions: Exercise, particularly multimodal combining aerobic resistance training, is cornerstone composition physical function SO. serves as an effective adjunct accelerating loss, strategies require integration exercise or prolonged implementation yield clinically meaningful Future research should prioritize standardized diagnostic criteria long-term assessments interventions.
