Exploring genetic diversity is essential for precise species delimitation, especially within taxonomically complex groups like passerine birds. Traditional morphological methods often fail to resolve boundaries; however, DNA barcoding, particularly through the mitochondrial cytochrome c oxidase subunit I (COI) gene, provides a powerful complementary method accurate identification. This study establishes comprehensive barcode library Iranian birds, analyzing 546 COI sequences from 94 across 23 families and 53 genera. There pronounced gap, with average intraspecific divergence at 0.41% interspecific 18.6%. Notable variation emerged in Persian nuthatch (Sittatephronota) Lesser whitethroat (Currucacurruca), while European goldfinch (Cardueliscarduelis) grey-crowned (Cardueliscaniceps) showed limited differentiation despite marked distinctions. Phylogenetic analysis revealed significant east-west splits C.curruca S.tephronota, reflecting Iran's geographic zoogeographic boundaries. These findings demonstrate effectiveness of barcoding elucidating biogeographic patterns, emphasizing key role as an ornithological crossroads avian biodiversity. Moreover, our results suggest that much gene arises synonymous mutations, highlighting purifying selection shaping mtDNA species.
