Abstract The plastid-targeted transcription factor Whirly1 ( WHY1 ) has been implicated in chloroplast biogenesis, plastid genome stability, and fungal defense response, which together represent characteristics of interest for the study autotrophic losses across angiosperms. While gene loss nuclear genomes well studied mycoheterotrophic plants, evolution molecular mechanisms impacting stability are completely unknown. Here we characterize four early-transitional orchid species genus Corallorhiza by synthesizing results phylogenetic, transcriptomic, comparative genomic analyses with sequences sampled from 21 orders We found an increased number non-canonical isoforms assembled all but greenest species, including intron retention some isoforms. Within , phylotranscriptomic revealed presence tissue-specific differential expression only most photosynthetically capable a coincident increase fully species. Gene- codon-level tests selective regimes did not infer significant signal either relaxed selection or episodic diversifying so late-stage full orchids Epipogium aphyllum Gastrodia elata . Taken together, this provides comprehensive view angiosperms to date. Our reveal that splicing alteration decreased deceased plastome group these changes may precede shifts observed

Abstract The Cucurbitaceae family includes a wide range of economically important fruits and vegetables; however, the laborious highly inefficient genetic transformation efficacy cucurbits has hindered exploration their gene functions. Virus-induced silencing (VIGS) technology, employed from antiviral RNA defense, emerged as viable alternative for high-throughput study plant function. In this study, we successfully established VIGS system utilizing Trichosanthes mottle mosaic virus (TrMMV), new member genus Tobamovirus . We demonstrated high durability mediated by TrMMV-VIGS vector in Nicotiana benthamiana , well several cucurbit species, including Cucurbita pepo Cucumis sativus C. lanatus melo insertion 90–400 bp fragments into led to effective target both with notably higher efficiency observed Furthermore, induced pronounced photobleaching phenotype flowers underscoring its potential application functional genomic research concerning floral traits particular species. Taken together, developed herein will facilitate rapid identification functions crops.

Auxin is an important class of plant hormones that play role in growth development, biotic stress response, and viruses often suppress host auxin levels to promote infection. However, previous research on auxin-mediated disease resistance has focused mainly signaling pathway, the molecular mechanisms how pathogenic proteins manipulate biosynthetic pathway remain poorly understood. TCP a plant-specific transcription factors, which TCP17 member binds promoter YUCCAs , key rate-limiting enzyme for synthesis, promotes expression involved synthesis plants. In this study, we reported Tomato spotted wilt virus (TSWV) infection suppressed through its interaction with TCP17. Further studies revealed NSs protein encoded by TSWV disrupts dimerization TCP17, thereby inhibit transcriptional activation ability reducing content Consequently, interference inhibits response signal Transgenic plants overexpressing exhibit against infection, whereas knocking out were more susceptible Additionally, other RNA (BSMV, RSV TBSV) can also interact interfere dimerization. Notably, overexpression enhanced BSMV. This suggests plays crucial defense different types use viral target component

As a high-value oilseed crop, soybean [ Glycine max (L.) Merr.] is limited by various biotic stresses during its growth and development. Soybean mosaic virus (SMV) devastating viral infection of that primarily affects young leaves causes significant production economic losses; however, the synergistic molecular mechanisms underlying response to SMV are largely unknown. Therefore, we performed RNA sequencing on SMV-infected resistant susceptible lines determine mechanism resistance SMV. When clean reads were aligned G. reference genome, total 36,260 genes identified as expressed used for further research. Most differentially (DEGs) associated with found be enriched in plant hormone signal transduction circadian rhythm according Kyoto Encyclopedia Genes Genomes analysis. In addition salicylic acid jasmonic acid, which well known disease resistance, abscisic indole-3-acetic cytokinin also involved immune soybean. Ca 2+ signaling related DEGs plant-pathogen interaction negatively influence resistance. Furthermore, MAPK cascade was either or responses SMV, depending different downstream proteins. The phytochrome interacting factor-cryptochrome-R protein module MEKK3/MKK9/MPK7-WRKY33-CML/CDPK play essential roles based protein-protein prediction. Our findings provide general insights into regulatory networks have potential improve legume infection.
