Abstract Aim Lithospermum erythrorhizon and Pueraria lobata exhibit promising potential as cosmetic additives for mitigating skin barrier impairment induced by photoaging. Despite their potential, the precise mechanisms underlying protective ameliorative effects remain elusive. This study sought to assess reparative properties of extracts (LP) on UVB‐irradiated human keratinocytes (HaCaT cells) explore therapeutic LP a protection agent. Materials Methods Antioxidant activities were gauged through 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH), 2,2′‐azino‐bis(3‐ethylbenzothiazoline‐6‐sulfonic acid) (ABTS), reactive oxygen species (ROS) assays. The expression levels barrier‐related markers, encompassing metalloproteinases (MMPs) hyaluronidase (HYAL) scrutinized using enzyme‐linked immunosorbent assay (ELISA), reverse transcriptase (RT)‐PCR, Western blotting, with particular focus involvement transforming growth factor (TGF)‐β/Smad nuclear factor‐κB (NF‐κB) signaling pathways. Results revealed that effectively scavenges free radicals, diminishes ROS production in dose‐dependent manner, significantly attenuates UVB‐induced MMP‐1 MMP‐3 modulation hyaluronan synthase (HAS)2/HYAL1 axis HaCaT cells. Additionally, demonstrated enhanced TGF‐β activation, fostering procollagen type I synthesis, concurrently exhibited mitogen‐activated protein kinases (MAPK)/NF‐κB inactivation, thereby pro‐inflammatory cytokine release alleviating cellular damage. Conclusion In conclusion, observed constituents highlight its substantial biological shielding against photoaging, positioning it candidate both pharmaceutical applications.

Background: Cycloheximide (CXM), an antifungal antibiotic, causes impaired memory consolidation as a side effect partially by disturbing the activities of central catecholaminergic and cholinergic system. Some reports indicated that puerarin prevented impairment in various models rodents. However, protective effects on cycloheximide for have not yet been investigated. Methods: The CXM-induced memory-consolidation impairment, produced administration AF64A neurotoxin, were investigated using passive avoidance task rats. A combination transmitter receptor agonists antagonists was used to explore nervous system function. activity antioxidant defense systems neurotransmitter prefrontal cortex hippocampus assayed. Results: Systemic (25 50 mg/kg, i.p.) or (5 10 µg/brain, i.c.v.) attenuated 1.5 mg/kg CXM (s.c.) improvements blocked antagonists, 5-HT2 agonist, adrenergic antagonist. Puerarin (only at reversed alterations levels norepinephrine monoamines hippocampus. also increased antioxidant-defense-system hippocampus, which had decreased CXM. Conclusions: We suggested attenuating may be due decrease oxidative damage normalition function

Neurodegenerative diseases encompass a wide range of debilitating and incurable brain disorders characterized by the progressive deterioration nervous system's structure function. Isoflavones, which are naturally occurring polyphenolic phytochemicals, have been found to regulate various cellular signaling pathways associated with system. The main objective this comprehensive review is explore neuroprotective effects isoflavones, elucidate underlying mechanisms, assess their potential for treating neurodegenerative disorders. Relevant data regarding isoflavones impact on were gathered from multiple library databases electronic sources, including PubMed, Google Scholar, Web Science, Science Direct. Numerous genistein, daidzein, biochanin A, formononetin, exhibited potent properties against diseases. These compounds modulate neurotransmitters, in turn contributes ability protect neurodegeneration. Both vitro vivo experimental studies provided evidence neuroprotection involve interactions estrogenic receptors, antioxidant effects, anti-inflammatory properties, anti-apoptotic activity, modulation neural plasticity. This aims provide current insights into characteristics shed light therapeutic applications future clinical scenarios.

Background: Puerarin is an isoflavone compound isolated from the roots of a leguminous plant, wild kudzu. Various functional activities this in multiple diseases have been reported. However, effect and mechanism puerarin improving blood pressure remain non-elucidated. Purpose: The current study was designed to assess preventive effects on onset progression hypertension verify hypothesis that alleviates by inhibiting ROS/TLR4/NLRP3 inflammasome signaling pathway hypothalamic paraventricular nucleus (PVN) salt-induced prehypertensive rats. Methods: Male Dahl salt-sensitive rats were fed low NaCl salt (3% drinking water) for control (NS) group or 8% (HS) induce prehypertension. Each batch divided into two treated bilateral PVN microinjection with either artificial cerebrospinal fluid through micro-osmotic pump 6 weeks. mean arterial (MAP) recorded, samples collected analyzed. Results: We concluded significantly prevented elevation effectively alleviated increase heart rate caused high salt. Norepinephrine (NE) plasma also decreased upon chronic infusion. Additionally, analysis sample revealed pretreatment positive cells gene level TLR4 (Toll-like receptor 4), NLRP3, Caspase-1 p10, NOX2, MyD88, NOX4, proinflammatory cytokines PVN. NF-κBp65 activity, inhibited oxidative stress, inflammatory responses Conclusion: conclude via inhibition PVN, suggesting therapeutic potential prevention hypertension.
