Abstract Background The panel of fluid- and imaging-based biomarkers available for neurodegenerative disease research is growing has the potential to close important gaps in clinic. With this growth increasing use, appropriate implementation interpretation are paramount. Various feature nuanced differences strengths, limitations, biases that must be considered when investigating etiology clinical utility. Emerging evidence suggests that an important function of the sleeping brain is removal wastes and toxins from central nervous system (CNS) due to activation waste (BWRS). The meningeal lymphatic vessels (MLVs) are part BWRS. A decrease in MLV associated with Alzheimer’s Parkinson’s diseases, intracranial hemorrhages, tumors trauma. Since BWRS activated during sleep, a new idea now being actively discussed scientific community: night stimulation might be innovative promising strategy for neurorehabilitation medicine. Abstract INTRODUCTION Changes in sleep physiology can predate cognitive symptoms by decades persons with Alzheimer's disease (AD), but it remains unclear which characteristics predict and neurodegenerative changes after AD onset. METHODS Using data from a prospective cohort of mild to moderate ( n = 60), we analyzed non‐rapid eye movement spindles slow oscillations (SOs) at baseline their associations amyloid beta (Aβ) tau cognition 3‐year follow‐up. RESULTS Higher spindle SO activity predicted significant Aβ baseline, lower Disease Assessment Scale Cognitive Subscale (better performance) score, higher Mini‐Mental State Examination score 36 months. Spindles SOs mediated the effect phosphorylated 181 (pTau181)/Aβ42 on cognition, while pTau181/aβ42 moderated cognition. Even prior to the onset of prodromal stages Alzheimer's disease (AD), a constellation sleep disturbances are apparent. A series epidemiological studies indicate that multiple forms these associated with increased risk for developing mild cognitive impairment (MCI) and AD, even triggering at an earlier age. Through combination causal manipulation in humans rodents, as well targeted examination disturbance respect AD biomarkers, mechanisms linking beginning emerge.
