Recent proliferation and integration of tissue-clearing methods light-sheet fluorescence microscopy has created new opportunities to achieve mesoscale three-dimensional whole-brain connectivity mapping with exceptionally high throughput. With the rapid generation large, high-quality imaging datasets, downstream analysis is becoming major technical bottleneck for connectomics. Current computational solutions are labor intensive limited applications because exhaustive manual annotation heavily customized training. Meanwhile, data always requires combining multiple packages secondary development by users. To address these challenges, we developed D-LMBmap, an end-to-end package providing integrated workflow containing three modules based on deep-learning algorithms mapping: axon segmentation, brain region segmentation registration. D-LMBmap does not require achieves quantitative projectome in a single superior accuracy cell types all modalities tested.

Abstract While CNS microglia have well-established roles in synapse pruning during neurodevelopment, only a few studies identified for formation. These focused on the cortex and primary sensory circuits restricted developmental time periods, leaving substantial gaps our understanding of early functions microglia. Abstract Repetitive transcranial magnetic stimulation (rTMS) is an emerging treatment for brain disorders, but its therapeutic mechanism unknown. We developed a novel mouse model of rTMS with superior clinical face validity and investigated the neural by which accelerated intermittent theta burst (aiTBS) – first rapid-acting antidepressant protocol reversed chronic stress-induced behavioral deficits. Using fiber photometry, we showed that aiTBS drives distinct patterns activity in intratelencephalic (IT) pyramidal tract (PT) projecting neurons dorsomedial prefrontal cortex (dmPFC). However, only IT exhibited persistently increased during both subsequent depression-related behaviors. Similarly, stress-related loss dendritic spines on IT, not PT neurons, further demonstrating cell type-specific effects stimulation. Finally, chemogenetic inhibition dmPFC blocked antidepressant-like aiTBS. Thus, demonstrate linking rapid aiTBS-driven to circuit plasticity.

The basal forebrain (BF) plays a crucial role in modulating cortical activation through its widespread projections across the mantle. Previous anatomical studies have demonstrated that each region receives specific projection from BF. In this study, we examined BF cholinergic and non-cholinergic to retrosplenial cortex (RSC) anterior cingulate (ACC) using two retrograde tracers, Fast Blue (FB) Fluoro-Gold (FG), combination with choline acetyltransferase (ChAT) immunostaining rats. RSC ACC receive mainly medial part of horizontal limb diagonal band (HDB) vertical (VDB). main difference RSC, ACC, prelimbic (PL) is PL rostral half globus pallidus (GP), whereas stronger VDB septum (MS). As injection site shifts caudal strong GP weak MS/VDB neurons are reversed. Cholinergic make up similar proportion total both (37%) (33%) injections. Double tracer injections revealed small number double-labeled HDB. These findings indicate spatially overlapping differential BF, varying between subregions different rostrocaudal regions.
