ABSTRACT Central melanocortin signaling plays a critical role in maintaining energy homeostasis by regulating intake and expenditure, with impairment of this system closely related to metabolic diseases such as obesity. Among receptor subtypes, 4 (MC4R) is the primary mediator these effects within central nervous system. Accumulating evidence suggests that MC4R contributes stress‐induced disruptions feeding behavior homeostasis. However, precise neural mechanisms which stress alters activity remain incompletely understood. In study, we compared brain‐wide c‐Fos expression patterns induced two distinct paradigms: lipopolysaccharide (LPS)‐induced inflammatory restraint male mice, further examined involvement MC4R‐expressing (MC4R + ) neurons conditions. We found both stressors elicited activation brain areas associated responses well regulation. Notably, LPS‐induced stress, but not selectively activated amygdala (CeA) oval nucleus bed stria terminalis (ovBNST). These results highlight recruitment during acute offering novel insights into imbalance depending on stressor types.

Abstract Chronic stress is a physiological state marked by dysregulation of the hypothalamic–pituitary–adrenal axis and high circulating levels hormones, such as corticosterone in mice or cortisol humans. This dysregulated may result development mood disorders, but process which this occurs still unknown. The bed nucleus stria terminalis (BNST) serves an integration center for signaling therefore likely important area disorders. project utilized chronic variable mild (CVMS) paradigm to persistently 6 weeks, followed RNA‐Sequencing anterodorsal (ad) BNST electrophysiology corticotropin‐releasing hormone‐expressing cells adBNST. Our results show significant sex biases transcriptome adBNST well effects CVMS on specifically males. Female‐biased genes are related synaptic transmission, while male‐biased RNA processing. Stress‐sensitive males transmission synapse formation. Additionally, data showed that suppressed M‐current not females. Immunohistochemical (IHC) staining situ hybridization (ISH) mRNA were performed on post-mortem human samples. Quantitative IHC revealed elevated levels both MDD BD, but only men, not women.
