With 5-year survival rates remaining constant at 6% and rising incidences associated with an epidemic in obesity metabolic syndrome, pancreatic ductal adenocarcinoma (PDAC) is on track to become the second most common cause of cancer-related deaths by 2030. The high mortality rate PDAC stems primarily from lack early diagnosis ineffective treatment for advanced tumors. During past decade, comprehensive atlas genomic alterations, prominence specific pathways, preclinical validation such emerging targets, sophisticated model systems, molecular classification into disease subtypes have all converged illuminate drug discovery programs clearer clinical path hypotheses. A deeper understanding cancer cell biology, particularly altered metabolism impaired DNA repair processes, providing novel therapeutic strategies that show strong activity. Elucidation tumor biology principles, notably a complexity immune regulation microenvironment, has provided exciting framework reawaken system attack cells.
