Abstract We investigated the role and mechanism of hsa-HLA-DRB1 in development progression diabetic foot ulcers. High-throughput sequencing was performed on three normal trauma tissues ulcer tissues. The circRNAs with significant differences were identified. downstream miRNAs predicted by miRanda RNAhybrid databases, mRNAs TargetScan database. Validation CCK8, flow cytometry, trabecular scratch assay, tubule generation Western blot, dual luciferase RT-qPCR. identified 461 significantly different circRNAs, which 260 up-regulated 201 down-regulated. Compared to tissue, highly expressed hsa-HLA-DRB1/miRNA_12118/FLT-1 axis constructed. In vitro, we found that HLA-DRB1 overexpression inhibited cell viability, wound healing, formation, promoted apoptosis, enhanced FLT-1 expression HUVECs. upregulation may promote hsa - HLA DRB1 inhibit biological function endothelial cells targeting miRNA_12118 acting FLT − 1. Circular RNAs (circRNAs) are increasingly recognized as significant regulators in multiple cancers, such hepatocellular carcinoma (HCC), frequently affecting microRNA (miRNA) expression. The diagnostic and prognostic roles of circRNAs, specifically circ_0009910 circ_0027478, conjunction with miR-1236-3p, HCC, have not yet been fully investigated. In this pilot study, we assessed the expression levels circ_0009910, miR-1236-3p plasma samples from 100 patients diagnosed HCC 50 healthy controls through reverse transcriptase–quantitative polymerase chain reaction (RT-qPCR). performance was evaluated using receiver operating characteristic (ROC) curve analysis, correlations clinicopathological features were examined. Circ_0009910 circ_0027478 exhibited upregulation (p < 0.05), whereas demonstrated downregulation 0.05). accuracy (area under [AUC] = 0.90), effectively differentiating showing a correlation tumor size, metastasis, alpha-fetoprotein (AFP) Both positive features, an inverse correlation.
