Alzheimer's disease (AD) and disease-related disorders (ADRD) are progressive neurodegenerative diseases without cure. occurs in 2 forms, early-onset familial AD late-onset sporadic AD. Early-onset is a rare (~1%), autosomal dominant, caused by mutations presenilin-1, presenilin-2, amyloid precursor protein genes the other late-onset, prevalent evolved due to age-associated complex interactions between environmental genetic factors, addition apolipoprotein E4 polymorphism. Cellular senescence, promoting impairment of physical mental functions constituted be main cause aging, primary risk factor for AD, which results loss cognitive function, memory, visual-spatial skills an individual live or act independently. Despite significant progress understanding biology pathophysiology we continue lack definitive early detectable biomarkers and/or drug targets that can used delay development ADRD elderly populations. However, recent developments studies DNA double-strand breaks result release fragmented into bloodstream contribute higher levels cell-free (cf-DNA). This cf-DNA released from various cell types, including normal cells undergoing apoptosis necrosis elevated blood have potential serve as blood-based biomarker detection ADRD. This proposed review aims to shed light on the major genetic and epigenetic contributions ageing process longevity of individuals. In this context, we summarize state knowledge most important variants, their interactions with environment, in achieving a healthy lifespan. We also explore contribution lifestyle influence non-heritable environmental factors (i.e., epigenetics). Accordingly, discuss role inflammageing as one targets overcome morbidity mortality older people for maintenance ageing. more integrated view will display not only underlying mechanisms at play but invites reader rethink both our attitudes toward age.

Despite the success achieved in increasing average life expectancy, problems of improving quality elderly and old people, prevention therapy age-associated diseases geriatric syndromes are still relevant. The immune system plays a key role aging process, functioning which undergoes wide range changes, collectively called "immunosenescence". Aging cells is associated with shortening telomeric regions chromosomes, loss protein homeostasis (proteostasis), mitochondrial dysfunction, DNA damage, oxidative stress. Senescent marked by resistance to apoptosis, cell cycle arrest abnormal production proinflammatory cytokines, leading development chronic low-grade inflammation. In this case, effector mechanisms response depleted, accompanied low ability adequately respond an antigenic stimulus. Thymus atrophy, depletion bone marrow niches, myeloid bias hematopoiesis change ratio lymphocyte subsets occur. addition, functions neutrophilic granulocytes impaired, but nature their during immunosenescence has not been sufficiently studied, so additional studies needed establish granulocyte dysfunctions immunosenescence.

Introduction: Metabolic age (MA) is the difference between an individual’s actual and of their body based on physiological biological factors. It indicator that reflects a person’s physical state, regardless chronological age. Insulin resistance (IR) health disorder in which tissues exhibit reduced response to circulating glucose uptake stimulated by insulin. Objective: The aim this study evaluate association MA, determined through bioelectrical impedance analysis, risk IR, assessed using validated scales, cohort Spanish workers. Methodology: A descriptive cross-sectional was conducted 8590 workers assess MA set sociodemographic variables, habits, IR scales such as Triglyceride–Glucose Index (TyG Index), Score for Resistance (METS-IR), Single Point Sensitivity Estimator (SPISE). Results: All analyzed variables were associated with values, strongest associations observed scale values (OR 4.88 [95% CI 4.12–5.65] METS-IR, 4.42 3.70–5.15] SPISE, 3.42 2.97–3.87] TyG Index) activity. Conclusions: influenced age, sex, social class; habits smoking, activity, adherence Mediterranean diet; values.

This study investigates the relationship between physical activity (PA) levels and DNA methylation (DNAm)-predicted epigenetic clocks in a U.S. population sample (n = 948, mean age 62, 49% female). Eight were analyzed, revealing that higher PA significantly associated with younger biological ages across all indicators, strongest effects observed for SkinBloodAge LinAge. Multivariable linear regression models, adjusted sociodemographic lifestyle factors, highlighted potential of to reduce ageing. Subgroup analyses indicated these associations more pronounced among non-Hispanic whites, individuals BMI 25-30, former smokers, suggesting impact varies different groups. These findings emphasize role slowing ageing reducing age-related health risks. Promoting regular PA, especially older adults those BMI, could improve well-being lifespan, highlighting as modifiable factor healthy disease prevention.

Background: Physical activity is an important predictor of quality life in older adults with type 2 diabetes (T2D). Unfortunately, most T2D adopt a sedentary lifestyle. The randomized, controlled TRIPL-A trial aims to verify the effect personalized, discontinuous exercise program on lifestyle adults. Methods: A total 305 patients (mean age ± SD: 68.8 3.3 years) were divided into control arm receiving only behavioral counseling and intervention 18-month supervised (ERS). primary outcomes changes sitting time (ST) metabolic equivalent (MET) values, both evaluated by International Activity Questionnaire short form. repeated measures ANOVA Bonferroni correction for multiple comparisons was used compare study outcomes. Results: ST MET differed significantly during compared group (p = 0.028 p 0.004, respectively). In group, decrease from baseline at 6 months 0.01) increase values up 12 < found. No significant differences found other variables. Conclusions: Beneficial within first year intervention. These results align theory change.
