Polyphenols represent the most studied class of nutraceuticals that can be therapeutics for a large spectrum diseases, including cancer. In this study, we investigated first time antitumor activities polyphenol extract from Annurca apple (APE) in MDA-MB-231 triple negative breast cancer cells, and explored underlying mechanisms. APE selectively inhibited cell viability caused G2/M phase arrest associated with p27 phospho-cdc25C upregulation p21 downregulation. promoted reactive oxygen species (ROS) generation cells while it acted as antioxidant non-tumorigenic MCF10A cells. We demonstrated ROS represented primary step activity pretreatment N-acetylcysteine (NAC) prevented APE-induced arrest, apoptosis, autophagy. downregulated Dusp-1 induced significant increase JNK/c-Jun phosphorylation were both by NAC. Introduction: Prodrugs have been used to improve the selectivity and efficacy of cancer therapy by targeting unique abnormal markers that are overexpressed cells absent in normal tissues. In this context, different strategies exploited new ones being developed each year. Areas covered: review, an integrated view potential use prodrugs targeted is provided.
