Beim Oropharynxkarzinom unterscheidet man inzwischen abhängig davon, ob eine HPV-16-Infektion ursächlich vorliegt oder nicht, in 2 verschiedene Entitäten. Neuere Daten zeigen ein diverses Bild der Bedeutung und Prävalenz des Surrogatparameters p16 (Diskordanz) für tatsächliche HPV-16-Assoziation. Im Rahmen Präventionsmöglichkeiten kommt Impfung hohe dem HPV-Screening gesunder Menschen geringe zu.

Next to alcohol and tobacco abuse, infection with human papillomaviruses (HPVs) is a major risk factor for developing head neck squamous cell carcinomas (HNSCCs), leading 350,000 casualties worldwide each year. Limited therapy options drug resistance raise the urge alternative methods such as photodynamic (PDT), minimally invasive procedure used treat HNSCC other cancers. We prepared lipid-coated polymeric nanoparticles encapsulating curcumin photosensitizer (CUR-LCNPs). The CUR-LCNPs were in nanometer range (153.37 ± 1.58 nm) showed an encapsulation efficiency of 92.69 0.03%. Proper lipid coating was visualized using atomic force microscopy (AFM). tested three HPVpos HPVneg lines regarding their uptake capabilities vitro killing capacity, revealing variable but highly significant tumor inhibiting effect all lines. No differences detected between groups (mean IC50: (9.34 4.73 µmol/L vs. 6.88 1.03 µmol/L), suggesting CUR-LCNPs/PDT be promising therapeutic option patients independent HPV status.

Abstract Purpose: Our goal was to demonstrate that lymphatic drainage fluid (lymph) has improved sensitivity in quantifying postoperative minimal residual disease (MRD) locally advanced human papillomavirus (HPV)-associated oropharyngeal squamous cell carcinoma (OPSCC) compared with plasma, and leverage this novel biofluid for patient risk stratification. Experimental Design: We prospectively collected lymph samples from neck drains of 106 patients HPV (+) OPSCC, along 67 matched plasma samples, 24 hours after surgery. PCR next-generation sequencing were used quantify cancer-associated cell-free (cf-HPV) tumor-informed variants plasma. Next, cf-HPV TNM stage, extranodal extension (ENE), composite definitions high-risk pathology. then created a machine learning model, informed by MRD clinicopathologic features, compare progression-free survival (PFS). Results: Postoperative enriched (P < 0.0001) correlated pN2 stage = 0.003), ENE 0.0001), trial-defined pathologic criteria (mean AUC 0.78).
