Abstract Ferroptosis is a type of regulated cell death characterized by its non-apoptotic, iron-dependent and oxidative nature. Since discovery in 2012, extensive research has demonstrated pivotal roles tumorigenesis, metastasis cancer therapy. The tumor microenvironment (TME) complex ecosystem comprising cells, non-cancer extracellular matrix, metabolites cytokines. Recent studies have underscored new paradigm which cells the TME, such as immune stromal also play significant regulating progression therapeutic resistance typically through complicated crosstalk with cells. Notably, this TME were partially mediated ferrotopsis-related mechanisms. This review provides comprehensive systematic summary current findings concerning ferroptosis how ferroptosis-mediated reprogramming impacts progression. Additionally, outlines various ferroptosis-related strategies aimed at targeting TME.

Abstract Pancreatic cancer remains one of the most lethal diseases worldwide. Cancer-derived exosomes, benefiting from protective role lipid membrane, exhibit remarkable stability in circulatory system. These released by tumor microenvironment, contain various biomolecules such as proteins, RNAs, and lipids that plays a pivotal mediating distant communication between local pancreatic other organs or tissues. They facilitate transfer oncogenic factors to sites, contributing compromised body immune system, metastasis, diabetes, cachexia, promoting microenvironment conducive growth metastasis patients. Beyond their intrinsic roles, circulating exosomes peripheral blood can be detected accurate liquid biopsy. This approach offers novel promising method for diagnosis management cancer. Consequently, are not only crucial mediators systemic cell-cell during progression but also hold great potential precise tools treatment. Exosome-based biopsy therapy represent advancements treatment Exosomes serve drug delivery vehicles, enhancing targeting efficacy anticancer treatments, modulating facilitating gene editing suppress growth. Ongoing research focuses on biomarker identification, systems, clinical trials validate safety exosome-based therapies, offering new possibilities early precision Leveraging therapeutic including ability deliver targeted drugs modulate responses, opens avenues innovative strategies.

Circular RNAs (circRNAs) have emerged as a prominent class of covalently closed single-stranded RNA molecules that exhibit tissue-specific expression and potential biomarkers in extracellular vesicles (EVs) derived from liquid biopsies. Still, their characteristics applications EVs remain to be unveiled. We performed comprehensive analysis EV-derived circRNAs (EV-circRNAs) using transcriptomics data obtained 1082 human body fluids, including plasma, urine, cerebrospinal fluid (CSF), bile. Our validation strategy utilized RT-qPCR immunoprecipitation assays, complemented by computational techniques for analyzing EV-circRNA features RNA-binding protein interactions. identified 136,327 EV-circRNAs various fluids. Significantly, considerable amount with high back-splicing ratio are highly enriched compared linear RNAs. Additionally, we discovered brain-specific plasma cancer-associated linked clinical outcomes. Moreover, demonstrated the serve evaluating immunotherapy efficacy non-small cell lung cancer (NSCLC). Importantly, involvement RBPs, particularly YBX1, sorting mechanism into EVs. This study unveils extensive repertoire across biofluids, offering insights disease mechanistic roles within The identification specific elucidation RBP-mediated mechanisms open new avenues application diagnostics therapeutics.

Abstract The morphogenesis of higher plants requires communication among distant organs throughout vascular tissues (xylem and phloem). Numerous investigations have demonstrated that phloem also act as a distribution route for signalling molecules being observed different macromolecules translocated by the sap, including nucleic acids proteins, change under stress situations. participation extracellular vesicles (EVs) in this has been suggested, although little is known about their role. Non-invasive diagnostic methods, including medical imaging techniques and blood biomarkers such as alpha-fetoprotein (AFP), have been crucial in detecting hepatocellular carcinoma (HCC). However, are only effective for tumor size larger than 2 cm. AFP measurement remains unsatisfactory due to high rate of misdiagnosis underdiagnosis. Therefore, new reliable better non-invasive approach necessary HCC identification.
