Background: Intrauterine fetal death and perinatal represent one of the most relevant medical scientific problems since, in many cases, even after extensive investigation, causes remain unknown. The considerable increase legal litigation obstetrical field that has witnessed recent years, especially cases stillborn births, simultaneously involved figure forensic pathologist research aimed at clarifying pathophysiological processes underlying stillbirth. Methods: our study aims to analyze sudden intrauterine unexplained syndrome (SIUD) evaluate role oxidative stress complex pathogenetic process In particular, immunohistochemical expression specific markers (NOX2, NT, iNOS, 8-HODG, IL-6) was evaluated tissue samples placentas SIUDs belonging case series (20 cases), collected from autopsy University Ferrara Politecnica delle Marche between 2017 2023. Results: demonstrated involvement deaths placenta examined. SIUD, expressed were NOX2 8-HODG. Conclusions: contributes investigating modulating different pathways tissues.

Pregnancy at advanced maternal age (AMA) is a condition of potential risk for the development maternal-fetal complications with possible repercussions even in long-term. Here, we analyzed changes plasma redox balance and effects on human umbilical cord mesenchymal cells (hUMSCs) AMA pregnant women (patients) various timing pregnancy. One hundred patients twenty younger than 40 years (controls) were recruited, evaluated timings during pregnancy until after delivery. Plasma samples used to measure thiobarbituric acid reactive substances (TBARS), glutathione nitric oxide (NO). In addition, was stimulate hUMSCs, which tested cell viability, oxygen species (ROS) NO release. The obtained results showed that throughout delivery levels lower those controls, while TBARS higher. Moreover, reduced viability release, increased ROS release hUMSCs. Our highlighted alterations presence potentially harmful circulating factors They could have clinical relevance prevention related

Pregnancy involves extracellular vesicles (EVs) through mechanisms that are poorly understood to date. Furthermore, it is not surprising EVs may also be involved in the pathophysiology of pre-eclampsia (PE) and gestational hypertension, two clinical conditions with high morbidity mortality, given their capacity mediate intracellular communications regulate inflammation angiogenesis. We searched major online scientific search engines (PubMed, Google Scholar, Scopus, WES, Embase, etc.) using terms “Preeclampsia”, “Pregnancy”, “Hypertension”, “Pregnancy-related hypertension”, “Extracellular vesicles”, “Biomarkers”, “Gestation” AND “Obstetrics”. Finding potential early biomarkers risk or illness progression would essential for optimum care expectant mothers aforementioned conditions. Nevertheless, none various screening assays have been discovered recently shown predictive values. The analysis peripheral blood starting from first trimester pregnancy hold great promise possible correlation hypertension problems represent a marker stages disease. use novel therapeutic approach management illnesses, as well. In order define EVs’ function physiopathology pregnancy-associated PE, well tools, we compiled most recent data this review.

Extracellular vesicles (EVs) could be involved in the onset of systemic sclerosis (SSc) through modulation vascular function. Anyway, available data are contradictory, and further investigation would necessary to clarify this aspect. Here, we characterized circulating EVs isolated from SSc patients evaluated their effects on human endothelial cells (HUVECs) smooth muscle cells. In 13 complicated 27 uncomplicated five healthy controls (HCs), analyzed size, concentration, surface marker expression. addition, were used stimulate HUVECs, cell viability, mitochondrial membrane potential, nitric oxide (NO) reactive oxygen species (MitoROS) release. cells, calcium movement examined. The results showed that expressed markers T-lymphocyte/platelet/endothelial origin larger more concentrated than those HCs. reduced viability potential increased NO MitoROS release HUVECs intracellular conclusion, found a specific pattern for patients, which have pathogenic role direct actions

Chromogranin B (CgB) is involved in the control of cardiovascular system through regulation catecholamine release. Whether CgB can exert direct actions on endothelium has not yet been clarified. Here, we aimed to investigate effects cell viability, mitochondrial membrane potential, reactive oxygen species (ROS), glutathione (GSH), nitric oxide (NO) release, and cytosolic calcium concentration ([Ca2+]c) human vascular endothelial cells (HUVECs) cultured under both physiological peroxidative conditions. In HUVECs, experiments were conducted establish proper timing stimulation. Thereafter, specific assays used evaluate response HUVECs physiologic or oxidative stress conditions presence absence β-adrenergic receptor agonists antagonists intracellular pathways blockers. Analysis NO release revealed that was able cause increased Additionally, same analyses performed with H2O2, showed protective exerted by CgB, which also counteract ROS maintain GSH levels. Furthermore, played a dual role [Ca2+]c depending culturing conclusion, our data provide new information about function highlight its potential as agent against conditions, such those found diseases.
