Cross-reactive carbohydrate determinants (CCDs) are complex N-glycans shared among allergens of plant, insect venom, and nematode origin. In allergic humans, IgE anti-CCD often develop cause discrepancies between serological skin tests. Overall, CCD-IgE believed to be low pathogenic relevance. IgE-targeting CCDs also detected in companion animals, but their potential biological relevance unknown. Herein, we first establish that, 34 dogs with atopic dermatitis, the presence serum was 14 pets (41.2%). dogs, as heterogeneous, they differentially recognized four distinct CCD-expressing proteins. The associated a higher more frequent recognition plant extracts not intradermal Two different proteins did elicit immediate reactions when injected intradermally detectable IgE. Similarly, two induce activation mast cells passively transferred canine Altogether, these results suggest that likely have little blocking them allergen-specific tests is warranted avoid false-positive extracts.

Detecting IgE sensitizations in the serum of allergic dogs is commonly done using allergen extracts, but these are difficult to standardize. This article details development and validation Pet Allergy Xplorer (PAX; Nextmune, Stockholm, Sweden), first multiplex macroarray for detection sensitization extracts molecular components; PAX derived from (ALEX2; MacroArray Diagnostics, Vienna, Austria). The selection allergens, cartridge processing, strategy identifying blocking directed against cross-reactive carbohydrate determinants (CCD), method used determining positivity threshold described. included evaluations specificity its anti-IgE monoclonal antibody, binding target assay precision, internal consistency. Additionally, influence possible confounding factors, such as sample type, hemolysis, lipemia, bilirubinemia, elevated CCD-IgE, was tested. Finally, rates 23,858 European 145 environmental Hymenoptera venom allergens were summarized. accurate reproducible has a unique CCD-detection strategy; offer window on cross-reactivity.
