Multidrug antimicrobial resistance (AMR) represents a formidable challenge in the therapy of infectious diseases, triggered by particularly concerning gram-negative Enterococcus faecium, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, and Enterobacter spp. (ESKAPE) pathogens. Designated as "priority" 2017, these bacteria continue to pose significant threat 2024, during worldwide SARS-CoV-2 pandemic, where coinfections with ESKAPE members contributed worsened patient outcomes. The declining effectiveness current treatments against pathogens has led an increased disease burden increase mortality rates globally. This review explores sophisticated mechanisms driving AMR bacteria, focusing on Key bacterial contributing include limitations drug uptake, production antibiotic-degrading enzymes, alterations target sites, enhanced efflux systems. Comprehending pathways is vital for formulating innovative therapeutic strategies tackling ongoing posed resistant

Abstract Background This study aimed to investigate the population heterogeneity on carbapenem susceptibility in Klebsiella pneumoniae strains that acquired a bla OXA-232 -bearing ColE-type plasmid. Methods A plasmid was electroporated into two carbapenem-susceptible K. strains. High- and low-carbapenem-resistant subpopulations were identified isolated using patch plating. The subsequently subcultured antibiotic-free media, yielding distinct populations: stable, high-level carbapenem-resistant heterogeneous Antibiotic tests, time-killing assays, profiles conducted, along with competition assay performed growth curve analysis. To assess virulence, we human serum resistance Galleria mellonella infection measured expression of virulence genes qRT-PCR. Additionally, whole genome sequencing carried out for further anaysis. Results Introduction pOXA-232 resulted isogenic transformants profiles: an unstable, (HCR), highly virulent subpopulation; low-level (LCR), low-virulence subpopulation. Whole analyses revealed dysfunctionality ompK36 HCR subpopulations. Subculturing led re-emergence populations variations additional compensatory mutation 9,000 bp deletion genome. Thus, stable featuring both mutations developed. Conclusion demonstrated underlying can promote emergence antibiotic resistance, even introduction carrying gene, such as OXA-232. highlights critical need closely monitor bacterial dynamics.

Foodborne diseases are still a serious global health issue, especially in developing countries, this study aimed to encapsulate the essential oils of coriander ( Coriandrum sativum L.) nanoemulsion and explore its efficiency as an antioxidant antibacterial agent against foodborne pathogens. The oil (CNE) was fabricated using Tween 80 emulsifier. Ultrasonication applied for 8 min yield homogenous nanoemulsion. chemical profile investigated by gas chromatography/mass spectrometry (GC‐MS). physicochemical properties CNE were examined dynamic light scattering (DLS), UV‐visible (UV‐Vis) spectrophotometer, scanning electronic microscopic (SEM), atomic force microscope (AFM). assessed Escherichia coli Klebsiella pneumoniae implicated foodborne. GC‐MS analysis showed that linalool main component with 64.06%. biofabricted had Z‐average 147.5 nm zeta potential 1.9 mV. SEM images spherical. AFM revealed moderately flat surface. Interestingly, exhibited high agent; turn gives it be candidate natural active bio‐based preservative used tackle food poisoning besides adding value food.

Background:Acinetobacter baumannii is a significant pathogen and major contributor to healthcare-associated infections, particularly in intensive care units. Its high potential for developing multiple drug resistance (MDR) makes it challenging manage. This study investigates the prevalence patterns of MDR A. isolates over six-year period at university hospital Southern Italy. Objective: The aims this are evaluate recent trends baumannii, analyze patterns, assess impact antimicrobial diagnostic stewardship program implemented 2018. Methods: retrospective cohort was conducted University Hospital Campania "Luigi Vanvitelli" from 2018 2023. A total 191 blood, urine, wound samples were analyzed. Antimicrobial susceptibility testing performed following EUCAST guidelines. strains assessed across three periods: pre-pandemic (2018-2019), during pandemic (2020-2021), post-pandemic (2022-2023) Results: Among isolates, 89.5% classified as MDR. highest number occurred 2020, with blood cultures urine increasing by 40.9% 62.5%, respectively, while decreased 34.2%. implementation programs correlated reduction carbapenem 2020 2022. However, meropenem colistin persisted. 60.4% decline isolation 2023 suggests effective infection control measures. Conclusions: remains threat healthcare. Although there have been slight reductions interventions, persistent last-line antibiotics underscores urgent need alternative treatments, enhanced surveillance, stricter strategies.

Klebsiella (K.) pneumoniae is a major reason of multidrug- resistant infections worldwide. The Enterobacterales that produce β-lactamases represent significant sources within healthcare environments. This prevalence can be attributed to the challenges they pose in treatment, their high potential for transmission, and substantial economic personal impact. Recent studies highlight emergence multidrug-resistant K. strains which show resistance colistin, last-line antibiotic, arising from mutational inactivation mgrB regulatory gene. bacteria greatest clinical relevance are those with third fourth generation cephalosporins, extended spectrum ampicillin-cephalosporin. Carbapenems category antibiotics remain one limited options addressing caused by multi-drug pathogens. Typically, these reserved as final solution when other therapies prove ineffective. Nevertheless, rising carbapenem-resistant presents considerable obstacle efficacy. Continuous monitoring advancement alternative therapeutic strategies essential tackle escalating challenge antibiotic resistance. However, rise poses health challenges. Notably, , pan-resistant bacterium, associated morbidity rates, particularly public settings. study reviews various aspects antibacterial resistance, focusing on genes present genome mechanisms through it develops Through analysis factors, we intend threat posed urgent requirement effective measures counteract it. Surveillance diagnostics vital controlling outbreaks enable early detection diseases, allowing timely interventions prevent further spread. Accurate laboratory confirming cases informing responses, thereby ensuring allocation resources during an outbreak. In this work, have carried out review antibiotics, found its genome, well involved. Finally, focus main causing hospital acquired last few years.
