Alzheimer’s disease (AD) is a widespread neurodegenerative disorder and one of the major challenges for public health. Despite extensive research, role microglia in AD remains complex dual. The aim this review to summarize most recent advances research regarding dual concerning both immunomodulation pathological progression by considering mechanisms activation microglia, effects on Aβ clearance, tau pathology, impacts due genetic variations microglial functions. Among these findings are status M1 M2 phenotypes, crucial that variants like TREM2 have modulating response microglia. This describes how modulation signaling pathway might be exploited therapeutically treatment underlines relevance personalized medicine approach.

Alzheimer's disease (AD) is a complex neurodegenerative disorder and the major cause of dementia. Amyloid-β (Aβ) tau aggregation, mitochondrial dysfunction, microglial dysregulation are key contributors to AD pathogenesis. Impairments in blood-brain barrier have unveiled contribution immune system, particularly B cells, pathology. crucial component adaptive immunity, exhibit diverse functions, including antigen presentation antibody production. While their role neuroinflammatory disorders has been well-documented, specific function lacks adequate data. This review examines dual cells humoral immunity modulating brain inflammation explores recent advancements passive active immunotherapeutic strategies targeting pathobiology. We summarize preclinical clinical studies investigating cell frequency, altered levels, implications neuroinflammation immunotherapy. Notably, demonstrate protective pathological roles AD, influencing neurodegeneration through antibody-mediated clearance toxic aggregates inflammatory activation inflammation. Passive immunotherapies Aβ shown potential reducing amyloid plaques, while emerging as promising strategies, requiring further validation. Understanding interplay between microglia, dysfunction critical unraveling Their nature progression underscores need for precise therapeutic interventions optimize immunotherapy outcomes mitigate effectively.
