Purpose This study examined the roles of nuclear factor erythroid 2-related 2 (NRF2) and programmed death ligand 1 (PD-L1) in colon carcinogenesis, underscoring on sex differences tumor location. Materials Methods A total 378 participants were enrolled from Seoul National University Bundang Hospital: 88 healthy controls (HC), 139 patients with colorectal adenoma (AD), 151 cancer (CRC). Quantitative real-time polymerase chain reaction (PCR), methylation-specific PCR, immunohistochemistry (IHC) performed utilizing samples normal mucosa HC group. Results NRF2 mRNA expression was higher CRC group than AD groups, decreased methylation groups. protein expression, as evaluated by IHC, increased groups relative to that PD-L1 remarkably These patterns consistent both males females. In sex- location-specific analyses, lower female male CRC. significantly females, particularly right-sided Moreover, females exhibited compared group, levels those at other locations. Conclusion Differences indicate site-specific carcinogenesis based sex,

Introduction: Gastric cancer (GC) is characterized by high incidence and poor survival rates. Crocin, a natural carotenoid from saffron, exhibits antioxidant, anti-inflammatory, anti-tumor properties. Ferroptosis, an iron-dependent cell death driven lipid peroxidation, plays critical role in progression potential therapeutic target. This study investigates whether crocin inhibits GC proliferation inducing ferroptosis explores its underlying mechanisms. Methods: employed vivo vitro models to assess crocin’s effects on proliferation, apoptosis, migration, invasion, ferroptosis. Pathway enrichment analysis was performed differentially expressed genes post-crocin treatment. Lentiviral vectors were used knockdown overexpress GGTLC2, exploring effects. The UCSC JASPAR databases predicted Nrf2 binding sites the GGTLC2 promoter. Molecular docking evaluated affinity for GGTLC2. Immunofluorescence nuclear-cytoplasmic fractionation assays analyzed expression localization. ChIP-qPCR determined Nrf2’s regulatory modulatory Results: results demonstrated that significantly inhibited invasion of cells while promoting apoptosis. Differentially following treatment predominantly enriched pathways associated with oxidative stress Crocin downregulated oncogene thereby suppressing simultaneously apoptosis revealed stable between suggesting may directly target modulate expression. Additionally, facilitated translocation nucleus cytoplasm. confirmed binds promoter region regulate expression, attenuated this interaction. Discussion: In conclusion, our findings suggest crocin, as promising compound therapy, inhibit through Nrf2/GGTLC2 signaling pathway, tumor initiation progression. provides novel insights into molecular mechanisms highlights agent GC.
