Intracerebral hemorrhage (ICH), a specific subtype within the spectrum of stroke disorders, is characterized by its high mortality and significant risk long-term disability. Cryptococcus neoformans (Cn) causes life-threatening meningoencephalitis in individuals with AIDS. Cn's polysaccharide capsule is mainly composed of glucuronoxylomannan (GXM) and plays a key role the dysregulation immunity, resistance to antifungal drugs, systemic dissemination, including CNS invasion. Although recent studies have begun elucidate involvement microglia cryptococcosis, our knowledge these resident phagocytes control cryptococcosis limited. We investigated microglial responses Cn infection effect active capsular production by comparing wild-type H99 acapsular mutant cap59 strains using CX3CR1-EGFP transgenic mouse stereotaxic intracerebral model. Microglia had difficulty combating infection. Active secretion material altered morphology distribution around cryptococcomas or fungal brain lesions. It also affected infiltration peripheral immune cells Moreover, RNA sequencing analyses supported importance modulation. Chemotaxis assays demonstrated that H99, especially GXM, impaired motility phagocytosis.
