Biclustering has emerged as a promising approach for analyzing high-dimensional expression data, offering unique advantages in uncovering localized co-expression patterns that traditional clustering methods often miss and thus facilitating advancements complex disease research other biomedical applications. However, state-of-the-art identify distinct at the expense of losing information about specific patterns, some which have been used to define cancer subtypes or reflect progression cellular processes. Additionally, these exhibit poor effectiveness noisy environments. To address limitations, we propose bucket trend-preserving (BTP) pattern, novel generalization existing patterns. And developed an algorithm, TransBic, extract significant biclusters BTP-patterns. Specifically, TransBic transforms problem into identifying common multipartite acyclic tournament subdigraphs shared by subsets digraphs derived from given matrix. Compared with prominent tools, demonstrates superior performance all non-row-constant especially under noise data fluctuations. Furthermore, successfully identifies most disease-related pathways type 2 diabetes (T2D), colorectal cancer, hepatocellular carcinoma, breast outperforming tools this regard. Different previous generalizations, BTP-patterns capture up-regulation down-regulation dynamics. Through targeted analysis T2D uncovers biological processes affected risk factors, extending application biclustering analysis.

Abstract Gestational diabetes mellitus (GDM) predisposes offspring to the development of obesity and type 2 diabetes. While GDM is studied in context maternal insulin resistance, consequences lean, sensitive women for health are unclear. This preclinical study investigated whether lean dams characterized by reduced secretion affects metabolic health. Lean was induced short-term 60% high-fat diet low-dose streptozotocin injections before mating mice. The control received only (HF) or low-fat (LF). Glucose homeostasis chow-fed offspring. resulted decreased birth weight, that resolved at postnatal day 15 (PN15). Defects in mitochondrial fatty acid β-oxidation (FAO) impair metabolic flexibility, which is an essential process for energy homeostasis. Very-long-chain acyl-CoA dehydrogenase (VLCADD; OMIM 609575) deficiency the most common long-chain FAO disorder presenting with hypoglycemia as a clinical manifestation. To prevent hypoglycemia, triheptanoin-a triglyceride composed of three heptanoates (C7) esterified glycerol backbone-can be used dietary treatment, since it metabolized into precursors gluconeogenesis. However, studies investigating effect triheptanoin on glucose homeostasis are limited. understand role gluconeogenesis pathophysiology defects, we injected VLCAD-deficient (VLCAD-/-) mice 13C3-glycerol presence and absence heptanoate (C7). The incorporation blood was higher VLCAD-/- than WT mice, whereas difference disappeared C7. result correlates 13C enrichment liver metabolites mice.
