Abstract Chronic liver injury induces cirrhosis and facilitates hepatocarcinogenesis. However, the effects of this condition on hepatocyte proliferation differentiation are unclear. We showed that rodent hepatocytes display a ductular phenotype when they cultured within collagenous matrix. This process involves transdifferentiation without emergence hepatoblastic features is at least partially reversible. During reaction in chronic diseases with progressive fibrosis, some hepatocytes, especially those adjacent to ectopic ductules, demonstrate transdifferentiation, but majority increased ductules originate from existing bile system undergoes extensive remodeling. In injury, weak sustained, most regenerative nodules composed clonally proliferating suggesting small fraction maintain their proliferative capacity injury. mouse hepatocarcinogenesis models, activate expression various fetal/neonatal genes, indicating these cells undergo dedifferentiation. Hepatocyte‐specific somatic integration oncogenes mice demonstrated may be origin for broad spectrum tumors through conclusion, phenotypic plasticity heterogeneity mature important understanding pathogenesis tumors.

The effectiveness of fucoxanthin (Fx) in liver diseases has been reported due to its anti-inflammatory and antifibrotic effects. Mesenchymal stem cells (MSCs)-based therapy also proposed as a promising strategy for fibrosis treatment. Recent studies have shown that the co-administration MSCs drugs demonstrates pronounced effect on fibrosis.

Fat storage and weight gain are dominant traits for hibernating mammals. However, excessive fat accumulation may cause liver damage. Here, we explore the lipid metabolic processes of Himalayan marmot (Marmota himalayana), a rodent species. We find that unsaturated fatty acid (UFA) content in food was consistent with large increase body mass marmots. Metagenomic analysis shows Firmicutes Bacterium CAG:110 plays synergistic role by synthesizing UFAs, which is demonstrated fecal transplantation experiments, indicating gut microbiome promotes marmots hibernation. Microscopic examination results indicate risk appears at maximum weight; however, function not affected. Upregulations UFA catabolism insulin-like growth factor binding protein genes provide an entry point avoiding injury.

Hepatocellular carcinoma (HCC), a prevalent solid of significant concern, is an aggressive and fatal disease with increasing incidence worldwide poor therapeutic outcomes. The etiology pathological progression non-alcoholic steatohepatitis (NASH) -related HCC multifactorial multistage. However, no single animal model can accurately mimic the full NASH-related poses challenges in transition other mechanism studies. Herein, novel conditional inducible wild-type human HRAS overexpressed mouse (HRAS-HCC mouse) was established 100 % morbidity death rate about one month under normal diet lifestyle. Advanced symptoms like ascites, thrombus, internal hemorrhage, jaundice, lung metastasis first mimicked mouse. In-depth features NASH- related were demonstrated by staining, biochemical tests typical marker gene detections. Murine anti-PD-1 sorafenib combined treatment effectively prolonged mice survival, which further confirmed accuracy reliability this model. We speculated that overexpression may initiate THBS1-COL4A3 axis to induce NASH severe fibrosis developed into based on Protein–protein interaction (PPI) network RNA sequencing analysis. Collectively, our study firstly duplicates natural sequential progressions murine very short period, providing reliable preclinical tool for therapeutics evaluation HCC.

Liver fibrosis occurs in response to chronic damage and inflammation the liver. Leaving untreated, it can lead decreased liver function eventually progress cirrhosis, a more advanced irreversible state of damage. Clinical investigations showed that disease associated with neurological symptoms including anxiety, depression, cognitive decline. However, few therapeutic options are available for treating related brain pathologies simultaneously. In this study, we aim find candidates target liver-brain axis. Gossypetin, flavonoid from sedum, shows promising capability CCl 4 -induced mouse model. Short term gossypetin administration is sufficient ameliorate impaired pre-existing fibrosis, suppress MKK3/6-p38 MAPK p53 activation, abolish activation hepatic stellate cells Kupffer cells. Although observe no neuronal loss mice do astrogliosis microglial certain regions, especially hippocampus. Brief also potential alleviating neuroinflammation these regions. These results suggest axis be candidate patients symptoms.
