β-Glucans derived from yeast are recognized as beneficial food additives. This study evaluated crude β-glucan extracts Pichia kudriavzevii DPUL-51-6Y, Kluyveromyces marxianus DPUL-F15, and Saccharomyces cerevisiae DPUL-C6 strains for their anticolitis potential. Chemical analysis revealed that was the primary component (71.88-78.47% purity). Notably, S. extract displayed superior thermal stability hydration capacity. In RAW264.7 macrophages, pretreatment at 100 μg/mL significantly reduced LPS-induced nitric oxide production pro-inflammatory cytokines by suppressing NF-κB signaling through reduction of p65 IκB-α while simultaneously activating Nrf2 AHR pathways. DSS-induced colitis BALB/c mice, oral administration β-glucans alleviated intestinal damage enhancing tight junction protein expression restoring gut microbiota composition, characterized an increased abundance Lactobacillus Prevotella. These effects were correlated with microbial metabolites, including indole-3-lactic acid, indole-3-β-acrylic tryptophol, short-chain fatty acids (acetic, propionic, butyric acids). Mechanistically, mitigated dual activation Nrf2/AHR pathways inhibition NF-κB. suggests yeast-derived plays a significant role in mitigating inflammatory response may alleviate ulcerative reshaping community metabolite profiles host tract.

Akkermansia muciniphila (A. muciniphila), known as a next-generation probiotic, has been widely recognized for its beneficial effects in various metabolic diseases. While there is not much research whether live or pasteurized A. different on intestinal health. In the present study, strain of was isolated from healthy individuals, with and named Timepie001 Timepie001+, respectively. They were administered to dextran sulfate sodium-induced ulcerative colitis mice investigate their influences host The results showed that prophylactic supplementation alleviates symptoms by retarding weight loss, preserving tissue structure, modulating inflammatory cytokines (TNF-α, IL-1β), enhance colonic mucosal barrier upregulating expression tight junction protein Claudin-1. Interestingly, better effect compared muciniphila. Moreover, can regulates gut microbiome maintain homeostasis. This provides theoretical support widespread application postbiotics food industry.

The total flavonoids of Periploca forrestii Schltr. (TFPF) exhibit notable anti-rheumatoid arthritis (RA) properties, highlighted by their inhibitory action on TNF-α. This study investigates the specific compounds TFPF responsible for TNF-α inhibition and delineates mechanism action. L929 cells luciferase-based assays were used to assess anti-TNF-α activity. Additionally, MH7A Wistar rats employed establish in vitro vivo models RA. Chemical chromatography, thermal proteome profiling (TPP), molecular docking, surface plasmon resonance (SPR), cellular shift (CETSA), drug affinity responsive target stability tests (DARTS), transcriptomic analysis potent molecules protein targets pathways. H&E staining, immunofluorescence ELISA, Western blot pharmacodynamic studies these molecules. mitigated injury inhibited luciferase expression, both which induced Among eight identified, quercetin exhibited most Quercetin improved reduced cell apoptosis rate treated with TPP revealed interaction between TNF-α, was verified SPR, CETSA, DARTS assays. Transcriptomic TNF signaling pathway, suggesting that may inhibit activation NF-κB pathway. suppressed phosphorylation IκBα p65, blocked p65 nuclear translocation, mRNA expression COX-2, INOS, IL-6, IL-1β. Moreover, decreased inflammation joint damage Quercetin, a compound TFPF, interacts inhibits activity thereby suppressing TNF-α/NF-κB pathway alleviating symptoms These findings position as promising inhibitor treatment
