To analyze the clinical characteristics and molecular biomarkers of adult T-cell lymphoblastic lymphoma (T-LBL) to identify prognostic factors, evaluate efficacy different chemotherapy regimens, providing a basis for optimizing treatment strategies T-LBL. A total 89 Patients aged 18-72 years with T-LBL, confirmed via histopathological examination lymph nodes, extranodal tissues, or bone marrow, were retrospectively included. Clinical data, details, mutational profiles collected. Prognostic factors assessed based on characteristics, safety two regimens compared. Descriptive statistics used disease spectrum. Most patients (84.00%) presented advanced (stages III-IV). Mediastinal invasion was observed in 63 (70.80%), 59 (66.30%) exhibited B symptoms. Bone marrow involvement occurred 19 (21.20%), bulky mediastinum (>10 cm) present 50 (56.18%). Mutations detected 29 patients, NOTCH1 being most frequently mutated gene, followed by PHF-6, JAK-1, JAK-3, IL-7R, TP53. The complete response (CR) rate 51.69%. 3-year overall survival (OS) progression-free (PFS) rates 74.9% 58.80%, respectively. Multivariate analysis identified female sex, lack CR, elevated lactate dehydrogenase (LDH) levels (>2× normal) as independent predictors poor OS (58.25%). Chemotherapy LDH levels, sex PFS (21.24%). mRNA expression-based stemness index (mRNAsi) has been used for prognostic assessment in various cancers, but its application lung adenocarcinoma (LUAD) is limited, which the focus of this study. Background Nicotinamide (NAM) metabolism fulfills crucial functions in tumor progression. Cellular senescence is a hallmark for cancers, particularly in lung adenocarcinoma (LUAD). This study developed risk model using signature genes LUAD patients.
