Medial arterial calcification is a chronic systemic vascular disorder distinct from atherosclerosis and commonly observed in patients with kidney disease, diabetes, aging individuals. We previously showed that NR4A3 (nuclear receptor subfamily 4 group A member 3), an orphan nuclear receptor, key regulator apo (apolipoprotein) A-IV-induced progression; however, its role poorly understood.

Abstract The negative effects of obesity on the cardiovascular health have drawn much attention. Weight-adjusted-waist index (WWI) has been proved to reflect weight-independent centripetal obesity. However, association between WWI and abdominal aortic calcification (AAC) not reported before. Using data from National Health Nutrition Examination Survey 2013–2014, we aimed determine relationship AAC in adults aged ≥ 40 years. was determined by dividing waist circumference square root weight. measured dual-energy X-ray absorptiometry quantified Kauppila scores. Severe (SAAC) defined as an score > 6. We utilized weighed multivariable logistic regression generalized additive model explore independent AAC. Threshold were further calculated two-piecewise linear model. 3082 participants enrolled our analysis, which 48.2% male. positively associated with scores (β = 0.34, 95% CI 0.05–0.63) exhibited a nonlinear SAAC. On left breakpoint (WWI 11.11), SAAC (OR 2.86, 1.40–5.84), while no such found right 1.07, 0.77–1.48). Our findings indicated that may serve simple biomarker US

Abstract Vascular calcification (VC) is recognized as a pathological vascular disorder associated with various diseases, such atherosclerosis, hypertension, aortic valve stenosis, coronary artery disease, diabetes mellitus, well chronic kidney disease. Therefore, it life‐threatening state for human health. There were several studies targeting mechanisms of VC that revealed the importance smooth muscle cells transdifferentiating, phosphorous and calcium milieu, matrix vesicles on progress VC. However, underlying molecular need to be elucidated. Though there no acknowledged effective therapeutic strategy reverse or cure clinically, recent evidence has proved not passive irreversible comorbidity but an active process regulated by many factors. Some available approaches mechanism provide promising prospects therapy This review aims summarize novel findings interventions VC, including role inflammatory responses, endoplasmic reticulum stress, mitochondrial dysfunction, iron homeostasis, metabolic imbalance, some related signaling pathways progression. We also conclude controversial in clinical practice channel blockers, renin–angiotensin system inhibitions, statins, bisphosphonates, denosumab, vitamins, ion conditioning agents.

Berberine (BBR), an isoquinoline alkaloid natural product, is isolated primarily from Coptis chinensis and other Berberis plants. BBR possesses various bioactivities, including antioxidant, anti-inflammation, anticancer, immune-regulation, antimicrobial activities. Growing scientific evidence underscores BBR’s substantial neuroprotective potential, prompting increased interest scrutiny. In this comprehensive review, we elucidate the attributes of BBR, delineate underlying molecular mechanisms, assess its clinical safety efficacy. The multifaceted mechanisms responsible for neuroprotection encompass attenuation oxidative stress, mitigation inflammatory responses, inhibition apoptotic pathways, facilitation autophagic processes, modulation CYP450 enzyme activities, neurotransmitter levels, gut microbiota composition. Furthermore, engages numerous signaling PI3K/Akt, NF-κB, AMPK, CREB, Nrf2, MAPK to confer effects. This review aims provide a knowledge base, stimulate broader discourse, facilitate advancements in application neuroprotection.

Mitophagy is a type of autophagy that can selectively eliminate damaged and depolarized mitochondria to maintain mitochondrial activity cellular homeostasis. Several pathways have been found participate in different steps mitophagy. plays significant role the homeostasis physiological function vascular endothelial cells, smooth muscle macrophages, involved development atherosclerosis (AS). At present, many medications natural chemicals shown alter mitophagy slow progression AS. This review serves as an introduction field for researchers interested targeting this pathway part potential AS management strategy.

The pathophysiology of vascular disease is linked to accelerated biological aging and a combination genetic, lifestyle, biological, environmental risk factors. Within the scenario uncontrolled artery wall processes, CKD (chronic kidney disease) stands out as valid model for detailed structural, functional, molecular studies this process. cardiorenal syndrome relates detrimental bidirectional interplay between cardiovascular system. In addition established factors, group patients subjected plethora other emerging such inflammation, oxidative stress, mitochondrial dysfunction, vitamin K deficiency, cellular senescence, somatic mutations, epigenetic modifications, increased apoptosis. A better understanding mechanisms through which uremic milieu triggers maintains early has provided important new clues on inflammatory pathways factors alike, altered behavior cells in arterial wall. Advances biology opens avenues novel pharmacological nutritional therapeutic interventions. Such strategies hold promise improve future prevention treatment not only but also elderly general population.

Cardiovascular diseases (CVDs) are the leading cause of mortality worldwide. Others and our studies have shown that mechanical stresses (forces) including shear stress cyclic stretch, occur in various pathological conditions, play significant roles development progression CVDs. Mitochondria regulate physiological processes cardiac vascular cells mainly through adenosine triphosphate (ATP) production, calcium flux redox control while promote cell death electron transport complex (ETC) related cellular response. Mounting evidence reveal stress-induced mitochondrial dysfunction plays a vital role pathogenesis many CVDs heart failure atherosclerosis. This review summarized functions cardiovascular system under (graphical abstract). Neurodegenerative diseases pose a growing global health challenge, with limited effective therapeutic options. Mitochondrial dysfunction, oxidative stress, neuroinflammation, apoptosis, and autophagy are common underlying mechanisms in these diseases. Thymol is phenolic monoterpene compound that has gained attention for its diverse biological properties, including antioxidant, anti-inflammatory, immunomodulatory activities. supplementation could provide potential neuroprotection improve cognitive deficits, depressant-like effects, learning, memory impairments rodents. Mechanistic investigations reveal the neuroprotective effects of thymol involve improvement mitochondrial inflammatory response. Several signaling pathways, apoptotic, NF-κB, AKT, Nrf2, CREB/BDNF pathways also involved. In this review, thymol, molecular mechanisms, safety, applications, current challenges toward development as agent were summarized discussed. We hope review provides valuable insights further promising natural product agent.

Atherosclerosis is a complex inflammatory process associated with high-mortality cardiovascular diseases. Today, there growing body of evidence linking atherosclerosis to mutations mitochondrial DNA (mtDNA). But the mechanism this link insufficiently studied. progression involves different cell types and macrophages are one most important. Due their high plasticity, can demonstrate pro-inflammatory pro-atherogenic (macrophage type M1) or anti-inflammatory anti-atherogenic M2) effects. These two types, formed as result external stimuli, differ significantly in metabolic profile, which suggests central role mitochondria implementation macrophage polarization route. According this, we assume that mtDNA causing disturbances play an internal trigger, leading formation M1 M2. This review provides comparative analysis characteristics function possible associations linked inflammation-based pathologies including atherosclerosis.
