Single microbial pathogens or host-microbiome dysbiosis are the causes of lung diseases with suspected infectious etiology. Metagenome sequencing provides an overview microbiome content. Due to rarity most granulomatous collecting large systematic datasets is challenging. Thus, single-patient data often can only be summarized visually. To increase information gain from a single-case metagenome analysis we suggest quantitative and qualitative approach. The 16S metagenomic results 7 patients pulmonary sarcoidosis were compared those 22 healthy individuals. From lysed blood, total DNA was extracted sequenced. Cleaned reads identified taxonomically using Kraken 2 software. Individual visualized Sankey diagram, Krona chart, heat-map. We five genera that exclusively present significantly enhanced in - Veillonella, Prevotella, Cutibacterium, Corynebacterium, Streptococcus. Our approach characterize blood composition diversity rare at individual level. Investigation unknown etiology, such as could enhance our comprehension their origin pathogenesis potentially uncover novel personalized therapeutics.

Objective: To analyze the associations of gut and circulating microbiota with vitamin D 3 (VD3), type I interferon (IFNI), systemic inflammation, clinical profiles in chronic spontaneous urticaria (CSU) patients. Methods: A total 36 CSU patients VD3 insufficiency (VDI; serum 25(OH)VD3 < 30 ng/mL) sex-, age-, body mass index–matched non-VDI were enrolled. Fecal bacteria identified through 16S rRNA sequencing, inflammation biomarkers assessed using ELISA kits. IFNI response was determined by measuring stimulatory activity on IFNI-stimulated element HEK293 cells vitro luciferase assays. Results: Higher urticarial score over 7 days (UAS7), higher frequency levocetirizine resistance, more severe proinflammation but weaker observed VDI than (all P 0.05). strongly positively associated level both groups ( 0.001). In healthy conditions, blood was considered a sterile environment until the development of new analytical approaches that allowed for detection circulating bacterial ribosomal DNA. Currently, debate exists on origin microbiota. According to advanced research using dark field microscopy, fluorescent in situ hybridization, flow cytometry, and electron so-called microbiota have been detected blood. Conversely, others reported no evidence common Then, it hypothesized may derive from distant sites, e.g., gut or external contamination samples. Alteration microbiota's equilibrium lead dysbiosis and, certain cases, disease. Blood can interact cells, especially monocytes engulf microorganisms T lymphocytes via spontaneous binding membranes. Moreover, LPSs, extracellular vesicles, outer membrane vesicles red cells reaching organs. This review aims describe composition individuals those disease conditions. Furthermore, special emphasis is placed interaction host better understand mechanisms.

Background: Sarcoidosis is a chronic inflammatory disease that can affect multiple organs. The aetiology of sarcoidosis not fully understood, but there increasing evidence the microbiome may play role. blood collection microorganisms live in bloodstream. Additionally, we aim elucidate potential causal relationship between these measures, phenomenon pathogenesis sarcoidosis. Conclusion: In present review, investigated alpha characteristics communities, examining indicators dysbiosis, probablemechanisms participation. A descriptive qualitative comparison conducted lung data patients healthy adults. This elucidates common taxa two microbiomes identifies potentially involved
