The human gut microbiota, consisting of trillions microorganisms, plays a crucial role in gastrointestinal (GI) health and disease. Dysbiosis, an imbalance microbial composition, has been linked to range GI disorders, including inflammatory bowel disease (IBD), irritable syndrome (IBS), celiac disease, colorectal cancer. These conditions are influenced by the interactions between host immune system, gut-brain axis. Recent research highlighted potential for microbiome-based therapeutic strategies, such as probiotics, prebiotics, fecal microbiota transplantation (FMT), dietary modifications, restore balance alleviate symptoms. Networks are widely used to represent relationships between objects, including microorganisms within ecosystems, based on high-throughput sequencing data. However, challenges arise with appropriate statistical algorithms, handling of rare taxa, excess zeros in compositional data, and interpretation. This work introduces a novel Slope-Matrix-Graph (SMG) algorithm identify microbiome correlations primarily slope-based distance calculations. SMG effectively handles any proportion data involves: (1) searching for correlated (e.g., positive negative directions changes) “target interest” setting, (2) quantifying graph changes via distances objects. Evaluations simulated datasets demonstrated SMG’s ability accurately cluster microbes into distinct positive/negative correlation groups, outperforming methods like Bray–Curtis SparCC both sensitivity specificity. Moreover, superior accuracy detecting differential abundance (DA) compared ZicoSeq ANCOM-BC2, making it robust tool analysis. A key advantage is natural capacity analyze zero-inflated without transformations. Overall, this simple yet powerful holds promise diverse analysis applications.
