The NOD-like receptor thermal protein domain associated 3 (NLRP3) inflammasome may play an important role in diabetic kidney disease (DKD). However, the exact link remains unclear. To investigate of NLRP3 DKD. Using datasets from Gene Expression Omnibus database, 30 inflammasome-related genes were identified. Differentially expressed selected using differential expression analysis, whereas intersecting based on overlapping differentially and genes. Subsequently, three machine learning algorithms used to screen genes, biomarkers identified by algorithms. Potential validated western blotting a db/db mouse model diabetes. Two biomarkers, sirtuin 2 (SIRT2) caspase 1 (CASP1), involved Leishmania infection pathway Both endothelial cells. Pseudo-temporal analysis cells showed that DKD mostly occurs during mid-differentiation stage. Western results CASP1 was higher group than control (P < 0.05), SIRT2 content decreased 0.05). provide potential theoretical basis for treatment.
