Background/Objectives: The study examined the relationships between specific Theory-of-Mind (ToM) dimensions, cognitive planning, and sleep duration in aging adults. Neurocognitive disturbances in Parkinson's disease (PD) are common and can impact several aspects of daily functioning quality life. This multicenter study evaluated the efficacy a novel immersive virtual reality (iVR) cognitive training (CT) targeting executive functions (EF)-planning, shifting, updating abilities-to improve prospective memory (PM) PD patients with mild impairment (PD-MCI). Potential transfer effects were also examined healthy older adults (HC). In this double-blind randomized controlled trial, 30 PD-MCI into 4-week EF-training (PD-CT) or active placebo (PD-AP), delivered at home through combined approach telemedicine iVR. Similarly, age- education-matched HC assigned to (HC-CT) (HC-AP). The PD-CT group exhibited significant improvements PM (in time-based verbal-response tasks) EF, particularly inhibition abilities Stroop test), sustained 2-month follow-up. These gains absent PD-AP group. Abstract INTRODUCTION The α‐synuclein (αSyn) seed amplification assay (αSyn‐SAA) is an accurate tool to detect αSyn seeds in patients with Parkinson's disease (PD). However, a minority of clinically diagnosed PD are negative for αSyn. METHODS αSyn‐SAA was performed cerebrospinal fluid (CSF) individuals ( n = 93), multiple system atrophy (MSA, 26), progressive supranuclear palsy (PSP, 18), corticobasal syndrome 3), and healthy controls 29). RESULTS detected 90% 81% MSA patients, while exhibiting high specificity (97%). SAA– had distinct phenotype compared SAA+ PD, including marked postural instability/gait disorder P 0.002), impaired episodic memory, lower CSF amyloid beta 42 0.03).
