Abstract Rheumatoid arthritis and osteoarthritis, the most common forms of arthritis, are chronic, painful, disabling conditions. Although both diseases differ in etiology, they manifest progressive joint destruction characterized by pathological changes articular cartilage, bone, synovium. While potent anti-inflammatory properties therapeutic (i.e., exogenous) glucocorticoids have been heavily researched widely used clinical practice, role endogenous susceptibility disease progression remains poorly understood. Current evidence from mouse models suggests that local glucocorticoid signaling is upregulated pro-inflammatory microenvironment rheumatoid aging-related mechanisms osteoarthritis. Furthermore, these indicate macrophages, mast cells, chondrocytes has effects, while fibroblast-like synoviocytes, myocytes, osteoblasts, osteocytes actions arthritis. Conversely, osteoblasts destructive actions. Together studies provide insights into pathogenesis inflammatory degenerative disease.

Gene-engineered immune cell therapies have partially transformed cancer treatment, as exemplified by the use of chimeric antigen receptor (CAR)-T cells in certain hematologic malignancies. However, there are several limitations that need to be addressed target more types. Natural killer (NK) a type innate represent unique biology surveillance. In particular, NK obtained from heathy donors can serve source for genetically engineered therapies. Therefore, NK-based therapies, including cells, CAR-NK and antibodies induce antibody-dependent cellular cytotoxicity emerged. With recent advances genetic engineering techniques, cell-based become promising approaches wide range cancers, viral infections, senescence. Accumulating clinical evidence indicates extensive inter-individual variations in the effectiveness and adverse effects of standard treatment protocols, which are largely attributed to multifactorial regulation hepatic CYP-dependent drug metabolism that is connected with either transcriptional or post-translational modifications. Age stress belong most important factors CYP gene regulation. Alterations neuroendocrine responses stress, associated modified hypothalamo-pituitary-adrenal axis function, usually accompany ageing. In this light, ageing followed by a decline functional integrity organs, including liver, failure preserving homeostasis under increased morbidity susceptibility among others, holds determinant role CYP-catalyzed thus, outcome toxicity pharmacotherapy. Modifications metabolizing capacity liver age have been reported particular, activity main isoforms male senescent rats, indicating decreased higher levels drug-substrates their blood. These along restricted experience use medicines childhood elderly, could explain at an extent variability efficacy outcomes, underscore necessity designing accordingly.

Age-associated osteoporosis (AAOP) poses a significant health burden, characterized by increased fracture risk due to declining bone mass and strength. Effective prevention early treatment strategies are crucial mitigate the disease burden associated healthcare costs. Current therapeutic approaches effectively target individual contributing factors AAOP. Nonetheless, management of AAOP is complicated multitude variables that affect its development. Main intrinsic extrinsic reviewed here, including mechanical unloading, nutrient deficiency, hormonal disbalance, disrupted metabolism, cognitive decline, inflammation circadian disruption. Furthermore, it discussed how these can be targeted for treatment. Although valuable as targets intervention, interconnectedness result in unique etiology every patient. Acknowledgement multifaceted nature will enable development more effective sustainable strategies, based on holistic, patient-centered approach.

Obesity, chronic pain, and aging are prevalent global challenges with profound implications for health well-being. Central to these processes adrenal hormones, particularly cortisol dehydroepiandrosterone (DHEA), along its sulfated form (DHEAS). Cortisol, essential stress adaptation, can have adverse effects on pain perception when dysregulated, while DHEA/S possess properties that may mitigate effects. This review explores the roles of in contexts obesity, acute aging, age-related diseases. We examine hormonal balance, specifically cortisol-to-DHEA ratio (CDR), as a key marker system functionality impact sensitivity, neurodegeneration, physical decline. Elevated CDR decreased levels associated worsened outcomes, including increased frailty, immune dysfunction, progression conditions such osteoporosis Alzheimer’s disease. synthesizes current literature highlight complex interplay between hormones their broader health. It aims provide insights into potential future therapies improve management promote healthy weight aging. By investigating mechanisms, this work contributes deeper understanding physiological intersections endocrine system.

Introduction: Osteoporosis is increasingly prevalent in the elderly, with fractures mostly occurring women and men who are older than 55 65 years of age, respectively. The aim this review was to examine evidence regarding influence hormones on bone metabolism, followed by clinical data hormonal changes attempt provide possible poorly explored diagnostic therapeutic candidate targets for management primary osteoporosis aging population. Material methods: An extensive Medline search using PubMed, Embase, Cochrane Library performed. Results: While rise Thyroid-stimulating hormone (TSH) levels has a protective role mass, decline estrogen, testosterone, Insulin-like growth factor 1 (IGF1), vitamin D cortisol, parathyroid hormone, follicle-stimulating (FSH) favor loss elderly. Particularly, AA rs6166 FSH receptor (FSHR) genotype, encoding more sensitive FSHR that encoded GG one, associated low total body mass density (BMD), independently circulating estrogen. A polyclonal antibody FSHR-binding sequence against β-subunit murine seems be effective ameliorating ovariectomized mice. Conclusions: complete assessment should completed both during evaluation. Novel tools might developed male female osteoporosis.

Abstract Background The Ecuadorian earthquake in April 16th was the second strongest and deadliest 2016 worldwide, with approximately one million people affected. In this paper, we analyzed psychological impact relationship between mental health events various earthquake-related stressors related to earthquake, 9 months after event. Methods We conducted an analytical cross-sectional study, applying anonymous survey 316 adolescents (13–19 years old) from Muisne, Ecuador. Suicidal tendency, depression, anxiety post-traumatic stress (PTSD) were evaluated via Child PTSD Symptom Scale (CPSS), Spence Children’s Anxiety , Okasha Suicidality Center for Epidemiologic Studies Depression (CES-D) adapted seven-questions survey. Results found a high prevalence of suicidal ideations behavior, posttraumatic stress, depression compared international studies. Even though currently living shelters had higher levels anxiety, their tendency significantly lower than those own or relatives’ home. Finally, not associated suicidality events, exception economic damage suffered by family. Conclusions High among high-school students found, especially who have serious damage. families unemployment rates parents seems be lack hope favorable perspectives future, situation that might lead emotional disturbances disorders. Although prolonged homelessness experience may stressful occurrence, also spiritual growth adolescents, work as protective factor against attempts.
