Background T follicular helper (TFH) cells, a subset of CD4 + Th play critical role in B cell activation, proliferation, and differentiation primarily within follicles secondary lymphoid organs, essential processes for effective antibody responses. TFH cells are also implicated various conditions, including autoimmune diseases, cancer, infectious allergies, vaccine reactions. Despite their broad impact, review the literature on tumors has not been conducted. We aimed to fill this gap by providing detailed analysis research landscape concerning tumors. Method conducted bibliometric from 2012 2024 using Web Science Core Collection (WoSCC). Abstract Objective This study assessed CLDN16 as a potential replacement or improvement biomarker for papillary thyroid cancer (PTC), addressing the limitations associated with prevalently used BRAF-V600E mutation. Design Database analyses, tissue validation, RNA sequencing, and functional assays were conducted to evaluate PTC its clinical application. Methods expression was examined in normal thyroid/para-tumor tissues compared across various types. We evaluated diagnostic accuracy, stability primary metastatic sites, associations aggressive features. Knockdown experiments performed investigate impact on cell behavior. Additionally, we developed support vector machine model diagnosing malignant high-risk PTCs. Results demonstrated high specificity PTC, positive detection rates (88.0% The Cancer Genome Atlas [TCGA] 88.3% our center) significantly surpassing (47.5% TCGA 74.3% center). resulted superior accuracy (ROC-CLDN16 = 0.922 vs ROC-BRAF-V600E 0.742 TCGA). exhibited stable sites features, including extrathyroidal extension lymph node metastasis. knockdown inhibited migration, invasion, iodine uptake cells. Clinically, effectively identified malignancy BRAF wild patients (94.2%), combined BRAF-V600E, achieved 96.9% accuracy. incorporation of into molecular typing facilitated precise identification (92.0% training set 100% validation set). Conclusions presents promising that could surpass offering effective utility revolutionizing identification.

γ-Glutamine cyclotransferase (GGCT) is an enzyme involved in the metabolic cycle of glutathione (GSH). Abnormal GSH metabolism mostly related to ferroptosis. However, mechanisms supporting aberrant GGCT expression PTC remain be investigated. In this study, we found that knockdown inhibited synthesis and promoted malondialdehyde (MDA) reactive oxygen species (ROS) accumulation, thereby promoting ferroptosis papillary thyroid cancer cells. Pro-GA, specific inhibitor GGCT, subcutaneous tumor formation K1 IP combined with LC-MS/MS showed interaction between RPS15A. RPS15A highly expressed tissues cells, promotes stability Knockdown p53 expression, which turn SLC7A11 resulting ferroptosis, while overexpression reversed GGCT-induced addition, miR-205-5p targeted 3' UTR inhibit GGCT-mediated growth, lung metastasis. conclusion, Mechanistically, interacts RPS15A, protein inhibits inhibiting synthesis. The upstream mechanism regulation by targeting GGCT.

In recent years, a growing body of evidence has suggested correlation between Hashimoto’s thyroiditis (HT) and the onset progression papillary thyroid carcinoma (PTC). However, mechanism underlying relationship HT PTC remains incompletely understood. This review discusses literature on summarizes research concerning immunological interplay these two conditions. It also delves into tumor‐associated cells (such as CD8+ T cells), macrophages (TAMs), regulatory (Tregs), cancer‐associated fibroblasts (CAFs), alongside other factors, including interleukins (ILs), interferon‐gamma (IFN‐ γ ), tumor necrosis factor‐alpha (TNF‐ α cyclooxygenase‐2 (COX‐2), inducible nitric oxide synthase (iNOS), hypoxia‐inducible factor‐1 (HIF‐1), highlighting their roles in interaction HT. We explore strategic direction immunotherapy malignancies, particularly with HT, propose novel targeted immunotherapies for advanced cancer.

Abstract Thyroid cancer is one of the most common types worldwide. It a spectrum different diseases, ranging from very indolent to lethal tumors. Differentiated Carcinoma (DTC), thyroid malignancy, has often an excellent prognosis, but some patients develop metastatic Radioiodine-Refractory disease (RAIR) that cannot be controlled locally. In this setting, and for with Medullary (MTC) Anaplastic (ATC), systemic treatment non-selective Multikinase Inhibitors (MKIs) employed improve survival rates quality life. The molecular characterization showed main drivers carcinogenesis not only correlate morphological clinical features can targeted by modern highly selective Kinase Inhibitors: vemurafenib dabrafenib carcinomas BRAF V600E mutation, including Papillary (PTC) its subtypes; in association MEK1/2 inhibitor trametinib V600E-mutant ATC; larotrectinib entrectinib NTRK fusions selpercatinib pralsetinib MTC RET point mutations DTC -fusions. Apart those markers, Microsatellite Instability status (MSI), Tumor Mutation Burden (TMB) PD1/PD-L1 assessment have been explored tumors, although immunotherapy ATC shown modest results. Herein, we present comprehensive review relevant markers predictive value pathology.
